Peran Maltodekstrin dan Sodium Kaseinat sebagai Enkapsulan terhadap Sifat Fisik dan Fisiologis Starter Yoghurt Mikroenkapsulas

Pengawetan starter bertujuan mempertahankan viabilitas dan sifat-sifat utama bakteri. Produksi kultur starter dengan pengeringan semprot belum dikembangkan karena rendahnya viabilitas. Tujuan penelitian adalah nlengkaji kemampuan maltodekstrin dan sodium kaseinat sebagai enkapsulan kombinasi pada mikroenkapsulasi starter yoghurt, dengan viabilitas tinggi, sebagai inokulum pada pembuatan yoghurt dan masa simpannya lebih lama. Metode penelitian menguji sifat mikrokapsul basil dari campuran maltodekstrin dan sodium kaseinat dalam proses mikroenkapsulasi menggunakan spray-drier, menguji ketahanan Streptococcus thermophilus dan Lactobacillus bulgaricus terhadap panas, melakukan mikroenkapsulasi masing-masing bakteri dengan mikrokapsul kombinasi, menguji kemampuan starter mikroenkapsulasi pada pembuatan yoghurt dan menguji lama simpan starter kering mikroenkapsulasi. Hasil penelitian menunjukkan perlakuan terbaik adalah mikrokapsul dengan Aw 0,28, kadar air 5,6%, kelarutan 98,1% dan mikrostruktur permukaan mikrokapsul tidak pecah/retak. Sel yang dipre-inkubasi 42°C, dipanen awal fase stasioner (S.thermophilus), dan fase stasioner ~Lb. bulgaricus) penurunan log jumlah sel lebih kecil 0,04 dan 0,05 dibanding 37 C, dipanen fase stasioner (S.thermophilus) dan fase log (Lb.bulgaricus) 0,23 dan 0,13; jumlah asam lemakjenuh meningkat dari 42,53% menjadi 44,42% pada S.thermophilus dan dari 12,90% menjadi 17,39% pada Lb.bulgaricus. Tingginya kerentanan bakteri mikroenkapsulasi dapat dikurangi dengan menyimpan pada suhu dan kelembaban yang rendah. Penurunan log jumlah sel mikroenkapsulasi terendah 0,29 terjadi pada konsentrasi sel sebelum pengeringan 1010 dibanding 0,56 dari konsentrasi 1011. Kualitas yoghurt dari starter mikroenkapsulasi tidak berbeda dengan kualitas yoghurt dari starter komersial. Penurunan log jumlah sel mikroenkapsulasi adalah terkecil 0,5 dan aktivitasnya terbesar (asam laktat 0,87%) serta paling stabil (8 bulan penyimpanan) pada suhu-kelembaban ruang penyimpanan 4°C-23%. Kesimpulan dari penelitian ini adalah enkapsulan kombinasi maltodekstrin dan sodium kaseinat dapat menstabilkan bakteri dengan kemungkinan didasarkan pada interaksi langsung maltodekstrin dengan membran bakteri maupun sodium kaseinat serta efek perlindungan dari sodium kaseinat dan meningkatnya resistensi kedua bakteri terhadap panas berkaitan dengan meningkatnya kandungan Saturated Fatty Acid dan menurunnya Unsaturated Fatty Acid Sensitivitas yang tinggi dari bakteri mikroenkapsulasi dapat dikurangi dengan menyimpan pada suhu dan kelembaban rendah.

The aim of preserving starter culture is to maintain bacteria viability and its major properties. The production of starter culture by a spray drying method has not been developed due to its low viability. This study was aimed to find out the capability of combination of maltodextrin and sodium caseinate as encapsulant for the microencapsulation of yoghurt starter with high viability as inoculum in the production of yoghurt with long shelf life. This experiment was carried out to study the properties of microencapsulant consisted of maltodextrin and sodium caseinate in the process of microencapsulation using spray-drier, to study the resistance of Streptococcus thermophilus and Lactobacillus bulgaricus to heat, to microencapsulate each bacteria with those combined microencapsulant, to study the capacity of microencapsulated starter on the processing of yoghurt and the storage life of dried microencapsulated starter. The results showed that the best treatment was the microcapsule with Aw value of 0.28, moisture content 5.6%, solubility 98.1% and no crack on the surface of the microcapsules. Cell which were pre-incubated at 42°C, harvested at the initial of stationary phase for S.thermophilus and stationary phase for Lb.bulgaricus showed lower reduction of log cell: 0.04 and 0.05 respectively compared to the one at 37°C, harvested at stationary phase for S.thermophilus, and log phase for Lb.bulgaricus: 0.22 and 0.13 respectively, whilst the fatty acids content increased from 42.53% to 44.42% for S.thermophilus and for Lb.bulgaricus from 12.90% up to 17.39%. The lowest amount of log cell reduction was 0.29 and it was found at cell concentration before drying 1010 compared to the one at cell concentration of 1011 was 0.56. There was no differens between yoghurt quality of yoghurt produced using microencapsulated starter and the one using commercial starter. The lowest reduction log cell of microencapsulated was 0,5 with highest activity (lactic acid 0,87%) and the most stable during storage of 8 months at 4°C with storage room humidity of 23% .. It can be concluded that the superiority of combination of maltodextrin and sodium caseinate as encapsulant in stabilizing dried bacteria is most likely due to the direct interaction of maltodextrin with bacterial membranes as well as sodium caseinate and on protective effect of sodium caseinate. Increasing resistance of bacteria to heat is associated with an increased content of SF A and a decrease in USF A. The higher susceptibility of dried microencapsulated bacteria in matrix was reduced by storing them at low temperatures and humidity.
