Pengaruh vitamin C terhadap proliferasi dan ekspresi vascular endothelial growth factor pada fibroblas normal

Rancangan penelitian yang digunakan adalah eksperimental sederhana, memakai kultur ï¬broblas kulit normal 1 orang relawan subkultur passage 3. Kultur ï¬broblas dibagi menjadi 6 keiompok, masing-masing kelompok perlakuan mendapat vitamin C 50 myug/ml, 100 myug/ml, 150 myug/ml, 200 myug/ml 300 myug/rnl dan kelompok tanpa perlakuan sebagai control. Selanjumya dilakukan pengukuran proliferasi ï¬broblas dengan spektrofotometer menggunakan MTT dan ekspresi VEGF ï¬broblas muggumkan ELISA. Perbedaan rerata proliferasi dan ekspresi VEGF fibroblast diuji dengan analisis varian satu jalan. Vitamin C terbukti dapat meningkatkan proliferasi fibroblas, namun tidak mampu meningkatkan ekspresi VEGF fibroblas normal.
The aim of this study is to determine whether vitamin C can be able to increase fibroblasts proliferation and increase VEGF expression by normal fibroblasts. A simple experimental study was conducted on 3rd passage normal human skin fibroblasts culture, isolated from one patient. Fibroblast cell culture was divided into 6 groups, each of treated group was given with vitamin C 50 myug/ml, 100 myug/ml, 150 myug/ml, 200 myug/ml, 300 myug/ml and untreated group as a control. Subsequently fibroblast proliferation was measured with spectrophotometer by using MTT-assay and VEGF expression by fibroblasts using ELISA. The mean differences of proliferation and VEGF expression by fibroblasts were analyzed using one-way Anova. Vitamin C is able to increase fibroblasts proliferation, however vitamin C can not increase VEGF expression by normal fibroblasts.
