Kualitas kontraksi (frekuensi, durasi, dan amplitudo) merupakan gambaran mekanik dari kontraksi yang dihasilkan oleh otot uterus. Perekaman dilakukan dalam keadaan spontan (tanpa diberi rangsang apa pun) dan dalam keadaan diberi oksitosin 0,01 IU pada organ bath yang di dalamnya terdapat sediaan otot uterus. Tujuan penelitian ini untuk mengkaji pengaruh air rendaman rumput fatimah (Anastatica hierochuntica L) terhadap kualitas kontraksi uterus tanpa oksitosin dan dengan perangsangan oksitosin. Penelitian dilakukan secara eksperimental laboratoris dengan rancangan post testonly control group design. Sampel penelitian adalah 40 sediaan otot uterus tikus rattus norvegicus galur Sprague dawley betina, umur 10-14 minggu, berat badan 150- 250 gram, berada pada fase estrus. Tikus dibagi menjadi 5 kelompok, kemudian perlakuan diberikan per oral berdasarkan kelompok, yaitu: kelompok kontrol (diberi air), kelompok pembanding (diberi estradiol), dan kelompok perlakuan yang diberi air rendaman rumput fatimah dengan variasi konsentrasi yaitu RF10, RF20 dan RF40. Masing-masing kelompok didekapitasi setelah 20 jam pemberian perlakuan per oral. Kemudian dilaparotomi dan diambil otot uterusnya. Selanjutnya dipasang pada water bath yang dihubungkan dengan kimograf dan kertas angus untuk direkam kontraksi yang terjadi tanpa dan dengan perangsangan oksitosin 0,01 IU. Data dianalisa dengan Anava satu jalur menunjukkan ada perbedaan signifikan hanya pada durasi (p=0,039) dan amplitudo (p=0,004) antara kelompok perlakuan dan kelompok kontrol pada kontraksi tanpa oksitosin. Uji t pada kelompok berpasangan menunjukkan ada perbedaan signifikan antara frekuensi kontraksi tanpa dan dengan perangsangan oksitosin 0,01 IU pada kelompok estradiol (p=0,032), RF10 (p=0,026), RF20 (p=0,001), dan RF40 (p=0,027). Juga ada perbedaan signifikan antara durasi kontraksi tanpa dan dengan perangsangan oksitosin 0,01 IU pada kelompok estradiol (p=0,005), RF10 (p=0.009), dan RF20 (p=0,005). Dapat disimpulkan bahwa pemberian air rendaman rumput fatimah per oral meningkatkan frekuensi dan mempersingkat durasi kontraksi otot uterus.

The quality of contraction (frequency, duration, and amplitude) is mechanical activities of uterine smooth muscle contractions. Recording was done without and with 0,01 IU oxytocin stimulation entered into organ bath within uterine smooth muscle strips. The aim of this research was to investigate the influence of water soaked of fatimah grass (Anastatica hierochuntica L) on quality of uterine smooth muscle contractions both in without and with 0,01 IU oxytocin stimulation. This research was laboratory experimental with post test only control group design. The samples were 40 uterine smooth muscle strips rattus norvegicus Sprague dawley 10- 14 weeks, 150 – 250 gr weight, on the estrous stages. The rats were divided into 5 groups. Control group was treatment by vehicle of solution (water), equivalent group was treatment by estradiol, and the others were treatment by fatimah’s grass 10 gr, 20 gr, and 40 gr that soaked into 350 cc hot water (70oC) during 12 hours. Twenty hours after treatment, rats were killed, and than uterine smooth muscle was removed and connected to chymograph to record the contractions. The data was analyzed by one-way Anova and the results showed that there was significant differences only in duration (p=0,039), and amplitude (p=0,004) between treatment groups and control or equivalent groups on the uterine smooth muscle strips without oxytocin stimulation. Paired t-test results showed that there was significant differences between frequencies without and with 0,01 IU oxytocin stimulation at estradiol (p=0,032), RF10 (p=0.026), RF20 (p=0,001), and RF40 (p=0,027) groups. Also there was significant differences between durations without and with 0,01 IU oxytocin stimulation at estradiol (p=0,005), RF10 (p=0,009), and RF20 (p=0,005) groups. It could be concluded that water soaked of fatimah’s grass increased the frequencies and shortened the durations of uterine smooth muscle contractions.
