Kadar testosteron serum dan spermatogenesis tikus putih dewasa (Rattus novergicus L.) setelah pemberian ekstrak daun wungu (Graptophyllum pictum L. Griff)

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun wungu (Graprophyllum pictum L. Griff) terhadap kadar testosteron serum, berat testis, indeks fertilitas,spermatogenesis, dan spermatozoa morfologi normal pada tikus jantan (Rattus novergicus.L). Lima belas ekor tikus jantan dan 45 ekor tikus betina berumur 9 – 10 minggu digunakan sebagai sampel penelitian. Lima belas ekor tikus jantan dibagi menjadi lima kelompok perlakuan dosis ekstrak daun wungu yaitu kontrol I (0mg/KgBB), kontrol II (0,5ml minyak zaitun), dosis 1 (100mg/KgBB), dosis 2 (200mg/KgBB), dan dosis 3 (400mg/KgBB) selama 60 hari. Perlakuan diberikan secara oral dan masing-masing kelompok terdiri atas tiga hewan uji sebagai ulangan Pada hari ke-55 sampai hari ke-60, masing-masing hewan uji dikawinkan dengan perbandingan satu jantan dikawinkan dengan tiga betina. Setelah 60 hari dilakukan pengamatan terhadap 15 ekor tikus jantan dan 45 ekor tikus betina untuk mengetahui kebuntingan. Pengamatan dilakukan terhadap kadar hormon testosteron serum dengan metode RIA (Radio Immuno-assay). Untuk menghitung lapisan sel spermatogenesis dibuat preparat histologi testis dengan metode paraffin, pewarnaan Hematoksilin-Eosin (HE), dan pengamatan morfologi spermatozoa normal dengan mikroskop. Data yang diperoleh diuji dengan Analysis of Variance Test (ANOVA a = 0,05) dan Duncan´s Multiple Range Test (DMRT) dilanjutkan dengan regresi linier. Berdasarkan pengamatan dan analisis statistik didapatkan hasil bahwa perlakuan dengan ekstrak daun wungu dengan dosis yang berbeda tidak berpengaruh nyata terhadap kadar testosteron, berat testis dan indeks fertilitas namun menunjukkan pengaruh nyata terhadap parameter jumlah sel spermatogonium, jumlah sel spermatosit, jumlah sel spermatid, jumlah total sel spermatogenik dan spermatozoa morfologi normal. Hal ini terlihat jelas dengan semakin besar dosis yang diberikan maka terjadi penurunan kualitas spermatogenesis dan spermatozoa morfologi normal. Berdasarkan analisis regresi linier terdapat korelasi nyata antara variasi dosis ekstrak daun wungu, indeks fertilitas, spermatogenesis dan jumlah normal spermatozoa. Namun, tidak terdapat korelasi nyata antara kadar testosteron terhadap berat testis. Hasil analisis kimia dengan menggunakan metode kromatografi gas menunjukkan bahwa ekstrak daun wungu mengandung senyawa steroid dan ß-sitosterol

This research objective is to identify the effects of wungu leaf extract (Graptophyllum pictum L. Griff) on the testosterone serum level, testis weight, fertility index, spermatogenesis and spermatozoa of normal morphology in male rat (Rattus novergicus.L). Fifteen male and 45 female rats of 9 to 10 weeks old were used as research samples. The fifteen male rats were divided into five treatment groups of wungu extract dosage: control I (0mg/KgBW), control II (0.5ml olive oil), dosage 1 (100mg/KgBW), dosage 2 (200mg/KgBW), and dosage 3 (400mg/KgBW) for 60 days. The extract was given orally and each group consisted of three test rats as repetition. On the day 55 to day 60, each test male animal was mated with three female rats. After 60 days, an observation was done to 15 male and 45 female rats to identify pregnancy. The serum testosterone level was assayed using RIA (Radio Immuno-assay) method. To count the spermatogenesis cell layers, a histological testis preparation was made with paraffin method, Hematoxylin-Eosin (HE), and normal spermatozoa morphological observation with microscope. The data obtained were tested by Analysis of Variance Test (ANOVA ? = 0.05) and Duncan’s Multiple Range Test (DMRT), followed by linear regression. Based on the observation and statistical analysis, it was found that wungu leaf extract of different dosages did not give significant effects on the testosterone level, testis weight and fertility index. However, it had significant effects on spermatogonium, spermatocyte, spermatide, total spermatogenic cell number, and spermatozoa of normal morphology. This was proven by the fact that the higher the dosage given, the worse the quality of spermatogenesis and spermatozoa of normal morphology. Based on Linear Regression Analysis, it was found significant correlation between dosage variation extract of wungu leaf and testosterone level, fertility index, spermatogenesis and number of normal spermatozoa. However, no significant correlation was found between testosterone level and testis weight. Chemical analysis results using gas chromatography indicated that wungu leaf extract contained steroid and ß-sitosterol compounds
