Pengaruh pemberian ekstrak air daun ceplikan (Ruellia Tuberosa L) terhadap kadar kreatinin dan ureum dalam serum serta gambaran histologis ginjal tikus putih (Rattus Norvegitus) diabetes mellitus

2008 | Tesis | S2 Ilmu Kesehatan MasyarakatLatar Belakang : Sampai saat ini, belum ada penelitian mengenai kemampuan hipoglikemik daun ceplikan (Ruellia tuberosa L). Daun ceplikan mengandung saponin, flavonoid, tanin 3,25 %, serat kasar 13,55 % dan seng (35,5 ppm) yang mempunyai manfaat sebagai antioksidan, komponen berbagai enzim, sintesis dan fungsi insulin. Kemungkinan juga mengandung senyawa xenobiotik yang bersifat toksik sehingga menyebabkan efek samping terhadap fungsi dan morfologis ginjal. Tujuan Penelitian: Mengetahui pengaruh ekstrak air daun ceplikan (Ruellia tuberosa L) terhadap kadar kreatinin dan ureum serum serta gambaran histologis ginjal tikus putih (Rattus novergicus) diabetes melitus. Metode Penelitian : Penelitian ini merupakan eksperimen murni dengan rancangan RCT dan dilaksanakan dari bulan Juni 2007 - Mei 2008. Subjek penelitian ini tikus putih (Rattus novergicus) jantan normal dan DM (induksi alloksan) sebanyak 54 ekor secara randomisasi. Data yang dikumpulkan yaitu kadar kreatinin dan ureum serum serta gambaran histologis ginjal. Pemberian ekstrak air daun ceplikan (Ruellia tuberosa L) dengan 3 variasi dosis. Pemberian glibenklamid 2,5 mg (kontrol positif) dan aquades (kontrol negatif). Gambaran histologis (metode pengecatan H&E ; PAS). Analisa data dengan menggunakan statistik non parametrik (p < 0,05) meliputi uji Kruskal Wallis dan Mann-Whitney . Hasil : Hasil penelitian menunjukkan bahwa ada pengaruh EADC terhadap penurunan ureum dan kreatinin tikus normal tetapi meningkat secara signifikan pada tikus DM dengan penurunan fungsi ginjal (p<0,050) ; Organ ginjal tikus DM mengalami kerusakan tubulus secara bermakna. Kesimpulan : EADC meningkatkan ureum dan kreatinin serum secara signifikan dan menimbulkan efek samping terhadap fungsi dan morfologis ginjal tikus DM dengan penurunan fungsi ginjal, sehingga daun ceplikan bisa digunakan oleh masyarakat sebagai obat tradional DM pada jalur non formal dengan terlebih dahulu melakukan pemeriksaan fungsi ginjal dan tetap memperhatikan pengaturan diit dan latihan jasmani secara teratur.
Background: Until today there is no study on hypoglicemic capacity of Ruellia tuberosa L which contains saponin, flavonoid, tanin 3.25%, gros fiber 13.55% and zinc (35.5 ppm). These have advantages as antioxidant, component of numerous enzymes, synthesis and insulin function. Ruellia tuberosa L may also contain xenobiotic compound that is toxic so that it may be have side effects to renal function and morphology. Objective: To identify the effect of Ruellia tuberosa L extract to level of creatinine and serum ureum as well as histologic description of the kidney of white rats (Rattus norvegicus) with diabetes mellitus (DM). Method: The study was an experiment with randomized control trial design and carried out from June 2007 to May 2008. Subject of the study more normal male white rats (Rattus norvegicus) and those with DM (alloxan included). There were as many as 54 samples randomly taken. Data collected consisted of level of creatinine and serum ureum as well as renal histologic description. Ruellia tuberosa L extract was given in 3 dosage variations and glibenclamide 2.5 mg as positive control , so aquades as negative control. Histologic description used H&E; PAS staining methode. Data analysis used non parametric statistics (p<0.050) comprising Kruskal Wallis and Mann-Whitney test. Result: The result of the study showed that there was significant effect of Ruellia tuberosa L extract to the decrease of ureum and creatinine of normal white rats but the effect was significant in diabetic rats with decreased to the function of kidney (p<0.050). Renal organ of white rats with DM had significant tubulus damage. Conclusion: Extract of Ruellia tuberosa L increase level of ureum and creatinin significantly and have side effects to the function and morphology of diabetic rats with decreased to the function of kidney.This result indicates that Ruellia tuberosa L could be use in society as traditional medicine for diabetes mellitus in a non-formal way and must be doing examination to the function of kidney and continuing exercise and diet.
