Patogenesitas Streptococcus iniae isolat dari Kupang pada ikan Patin (Pangasius sutchi)

Penelitian ini bertujuan untuk mengetahui patogenesitas S.iniae isolat dari Kupang, meliputi gejala klinis, perubahan histopatologik dan hematologik pada ikan patin (P. sutchi). Penelitian dilakukan dua tahap, pertama uji pendahuluan yang bertujuan untuk mengetahui LC50 S. iniae pada ikan patin. Sebanyak 50 ekor ikan patin ukuran 15 c m dibagi 5 kelompok, masing-masing 10 ekor. Kelompok 1 - 4 diinfeksi S. iniae dengan konsentrasi masing-masing 10 2, 10 4, 10 6, dan 10 8 sel/mL intramuskuler dan kelompok 5 tida k diinfeksi sebagai kontrol. Tahap kedua, dalam uji utama digunakan 35 ekor ikan patin terdiri dari: 30 ekor ikan diinfeksi S. iniae dengan konsentrasi 0,1 mL LC 50 intramuskuler dan 5 ekor ikan tidak diinfeksi, langsung diambil darahnya dan dinekropsi untuk pemeriksaan awal hematologik dan histopatologik (kontrol). Hari ke 2, 4 dan 6 masing-masing sebanyak 5 ekor ikan diambil darahnya untuk pemeriksaan hematologik dan 5 ekor ikan dinekropsi setiap hari untuk pemeriksaan histopatologik hingga hari ke 6 . Hasil uji pendahuluan diperoleh Nilai LC50 sebesar 6,79 x 106 sel/mL. Hasil uji utama menunjukkan, gejala klinis ikan patin yang diinfeksi S. iniae 0,1 mL 106 sel/mL: berenang lambat, kesulitan bernafas , refleks lambat, warna tubuh pucat, mulut dan pangkal sirip kemerahan. Jumlah lekosit total meningkat secara signifikan (P=0,05) pada hari ke 2, kemudian menurun pada hari ke 4 dan meningkat kembali secara normal pada hari ke 6 setelah infeksi. Jumlah monosit meningkat secara signifikan (P=0,05) pada hari ke 2, tetapi menurun terus hingga hari ke 6 setelah infeksi. Kongesti ditemukan pada hati, limpa, ginjal, dan otak pada hari ke 1 dan 2; nekrosis dan radang kulit hingga otot pada hari ke 3. Kulit dan otot melanjut dengan radang berdarah, serta radang usus pada hari ke 5. Nekrosis hati dan ginjal, kongesti dan radang ringan jantung pada hari ke 6 sesudah infeksi.

The research aimed to to know the patogenicity of S.Iniae isolat from Kupang, consist of the clinic symptom, change of histopatologic and hematologic at pangasius catfish (P. Sutchi). The research was done by two phase, the first test aimed to know LC50 of S. iniae at pangasius catfish. The 50 heads of pangasius catfish in 15 cm of length divided in 5 groups, each 10 heads . Group of 1 st to 4th were infectioned by S. iniae with each concentration: 10 2, 104, 106, and 108 cell/mL by intramuscular, and group 5th was not infectioned as a controll. Second phase, at t e primer test used 35 heads of pangasius catfish, h consist of: 30 heads were infectioned by S.iniae with concentration 0,1 mL LC50 by intramuscular, and 5th heads were not infectioned, but taken blood directly and necropted for the inspection early of hematologic and histopatologic (controll ). The 2nd, 4 th and 6th days, each 5 heads were taken blood for the inspection of hematologic and 5 heads were necropted everyday for the inspection of histopatologic up to 6 days. The result of first test was known equal to 6,79 x 106 sel/mL. The result of primer test indicate, the clinic symptom of pangasius catfish were infected by S. iniae with concentration 106 sel/mL consist of: swimming slowly, difficulty breathes, reflex slowly, pale body colour, reddish fin and mouth. Amount of total leucocyte was increase by significantly (P= 0,05) on to 2nd day, then rundown on to 4 th and increase again with normally on to 6 th day after infectioned. Amount of monosit increase by signifikan (P = 0,05) on to 2nd, but downhill contin ously till to 6th day after infectioned. Congesti were found at liver, spleen, kidney, and brain on to 1st and 2nd day; nekrosis and skin chafe till muscle on to 3rd day. Skin and muscle continued by chafed bleed, and also chafe intestine on to 5th day. Liver Nekrosis and kidney, kongesti and chafe lightly of heart on to 6 th day after infection.
