Peranan Gen Knati Arabidopsis thaliana dalam pembentukan tunas pada tanaman anggrek Phalaenopsis amabilis (L.) Blume transgenik

2007 | Tesis | S2 BiologiPenelitian ini bertujuan untuk mengetahui fungsi gen KNAT1 Arabidopsis thaliana sebagai transgen dalam pembentukan tunas pada tanaman anggrek Phalaenopsis amabilis (L.) Blume. Tanaman anggrek transforman 35S::KNAT1 diikuti perkembangan tunasnya. Karakter fenotip tanaman yang diamati meliputi bentuk daun, ukuran daun, dan ukuran akar. Keberadaan gen KNAT1 di dalam tanaman transforman ditentukan melalui amplifikasi fragmen 1,2 kb gen KNAT1 dengan teknik PCR. Hasil penelitian menunjukkan bahwa jumlah tanaman anggrek P. amabilis transgenik in vitro meningkat tinggi, yaitu sebesar 31,92 kali. Beberapa tanaman menunjukkan bentuk daun abnormal yang meliputi daun kasar (13,98%), daun beralur (1,69%), daun kasar multitunas (7,95%), daun terompet (0,72%), daun normal multitunas (13,97%), dan daun berlobi (14,93%). Data tersebut menunjukkan bahwa transgen KNAT1 menginduksi multiplikasi tunas dan perubahan bentuk daun pada tanaman anggrek P. amabilis transgenik. In order to understand the function of Arabidopsis thaliana KNAT1 gene as a transgene on shoot formation of Phalaenopsis amabilis (L.) Blume orchid plants, the shoot development of 35S::KNAT1 transformants was observed. Phenotypic features were observed including leaf shape, leaf size, and root size. The existence of KNAT1 gene in plants genome was confirmed through amplification of 1.2 kb fragment of KNAT1 gene by PCR technique. The results showed that the number of transgenic P. amabilis orchid plants increased highly, that was about 31.92 times. Abnormal leaf shapes were formed including rough leaf (13.98%), tangled leaf (1.69%), rough multishoot leaf (7.95%), trumpet leaf (0.72%), normal multishoot leaf (13.97%), and lobed leaf (14.93%). These data indicated that KNAT1 transgene might induced multiplication of shoot and alteration of leaf shape in transgenic P. amabilis plants.
