Pertumbuhan dan perkembangan embrio Rattus norvegicus L dari induk dengan perlakuan kurkumin pada fase organogenesis

Kurkumin merupakan zat warna kuning yang dikenal sebagai bahan alami yang memiliki aktifitas biologis diantaranya sebagai antiinflamasi, antikanker, antiinfeksi dan juga dikenal di masyarakat sebagai pengatur kesuburan pada wanita. Sampai saat ini kurkumin belum banyak diketahui efek terhadap pertumbuhan dan perkembangan pada embrionya. Tujuan penelitian disini untuk mengetahui perkembangan dan pertumbuhan embrio Rattus norvegicus L yang induknya diberi kurkumin selama fase organogenesis. Penelitian dilakukan terhadap 15 induk tikus Rattus norvegicus L, umur 2 - 3 bulan, berat 200 gram – 300 gram, dibuntingkan kemudian dibagi menjadi tiga kelompok, hari ke 6 sampai ke 15 diberi perlakuan, hari ke 18 didekapitasi. Kelompok I sebagai pembanding yang diberi larutan CMC (Carboxyl Methyl Cellulose), kelompok II sebagai perlakuan 1 yang diberi kurkumin 50 mg/Kg BB, dan kelompok III sebagai perlakukan 2 yang diberi kurkumin 75 mg / Kg BB setiap hari. Analisa digunakan regresi linier didapatkan hasil bahwa jumlah embrio pada kelompok pembanding lebih banyak dibandingkan dengan perlakuan 1 dan 2. semakin banyak dosis perlakuan kurkumin maka semakin sedikit jumlah embrionya. Panjang embrio pada perlakuan 1 menunjukkan peningkatan dibandingkan pada kelompok pembanding dan perlakuan 2. berat badan embrio menunjukan semakin banyak perlakuan maka semakin turun beratnya. Jumlah resorbsi embrio menunjukkan semakin banyak dosis perlakuan maka semakin banyakjuga jumlah resorbsi. Korteks adrenal embrio menunjukan semakin banyak dosis perlakuan maka semakin tipis korteks adrenalnya. Perlakuan 2 embrio menunjukkan adanya hipervaskularisasi pada kepala dan badan serta ditemukan adanya kecacatan pada tungkai depan.

Curcumine is a yellow material known as natural ingredient that has biological activities, like: anti- inflammation, anti-cancer and anti- infection. Besides, it is also popular in society as woman fertile regulator. However, the side effect of curcumine toward the growth and development of embryo is not well recognized. The objective of this research is therefore to study the growth and the development of dam’s embryo in curcumine treated Rattus norvegicus L at the organogenesis phase. The objects of this research were fifteen (15) rats Rattus norvegicus L, between 2-3 months of age and 200-300 gram in weight. These rats were allowed to mate to male fertile rat, then divided into three groups of five. The first group was treated with Carboxyl Methyl Cellulose (CMC). The two second groups were treated with curcumine at the dose of 50 mg and 75 mg / kg bw / day, respectively. CMC and Curcumine were later administered during 10 days, from 6th to 15th day of gestation. One day after treated animals were killed, uterus and embryos were dissected out. Embrios were weighted and fixed in 10% formalin, and the length of body was then measured. The adrenal of embryo were processed for histological observation, stained with Haematoxyline-Eosin to measure the thickness of adrenal cortex. Quantitative data were analyzed using linear regression. The result showed that (1) the total number of embryos were decreased if compared with CMC group. (2) the length of embryo increased in treated groups. (3) embryo in treated groups were found abnormal. (4) It is observed that the higher dose of curcumine the thinner the adrenal cortex. It was concluded that curcumine given at organogenesis phase possesses teratogenic effect.
