Pengaruh Biofosfonat risedronat per oral pada relaps gigi :: Penelitian experimental laboratoris pada Cavia Cobaya

2006 | Tesis | PPDGS I OrtodonsiaPenelitian ini bertujuan untuk mengetahui pengaruh bisfosfonat jenis risedronat yang diberikan secara oral pada gerakan relaps gigi marmut yang digerakkan dengan alat ortodontik. Subyek penelitian terdiri dari 30 ekor marmut yang sesuai dengan kriteria. Seluruh subyek dibagi dalam 5 kelompok, pada kelompok 1 adalah kelompok kontrol yang tidak diberikan bisfosfonat risedronat per oral, kelompok 2 diberikan bisfosfonat risedronat per oral dengan dosis 0,25 mg, kelompok 3 dengan dosis 0,50 mg, kelompok 4 dengan dosis 0,75 mg dan kelompok 5 dengan dosis 1 mg. Setelah dilakukan aklimatisasi pada marmut selama seminggu, dipasangkan alat ortodontik pada gigi insisivi atas berupa bonding cleat dan opencoil spring selama 21 hari agar terbentuk jarak interinsisivi sebagai jarak awal. Alat ortodontik tersebut dilepas setelah opencoil spring memanjang maksimal dan pada hari ke 21 tersebut mulai diberikan bisfosfonat risedronat per oral pada tiap kelompok sesuai dosisnya selama 7 hari berturut-turut. Perubahan jarak interinsisivi yang menandakan adanya relaps diukur setiap hari selama 7 hari. Pada hari terakhir diukur jarak interinsisivi sebagai jarak akhir. Selisih antara jarak awal dan akhir dihitung dan dilakukan analisis variansi (anava) untuk melihat perbedaan gerakan relaps antara kelompok kontrol dan kelompok pemberian bisfosfonat risedronat per oral. Dan untuk mengetahui perbedaan pengaruh antara masing-masing dosis obat risedronat terhadap kontrol digunakan uji post hoc. Hasil penelitian menunjukkan bisfosfonat risedronat per oral berpengaruh mencegah relaps gigi marmut setelah digerakkan dengan alat ortodontik dan terdapat perbadaan bermakna antara kontrol dan kelompok pemberian bisfosfonat risedronat per oral (p<0,05). Dosis minimal pemberian risedronat per oral dapat menghambat relaps gigi marmut adalah 0,50 mg dan sangat efektif pada dosis 1 mg.
The aim of this research was to found the effect of bisphosphonate risedronate which given to the movement of rabbits teeth relapse orally, which moved by the orthodontic utensils. Research subject consist of 30 rabbits which appropriate with the requirements. From the whole subjects were devided to 5 groups. Group 1 was the controlling group which have not been given bisphosphonate risedronate per oral. Group 2 have been given bisphosphonate risedronate per oral 0,25 mg dosage. Group 3 with 0,50 mg dosage, group 4 with 0,75 mg dosage and group 5 with 1 mg dosage. Orthodontic utensils were put on the incisivus teeth above, which shaped as bonding cleat and opencoil spring for about 21 days after done the aclimatisation to the rabbits for a week, so that it could make interincisivus gap as the first gap. Those orthodontic utensils were released after opencoil spring reached the maximum gap. Due on the 21th day started to give bisphosphonate risedronate per oral for each groups based on the dosage for 7 days continuously. The gap changes of interincisivus which shown relapse was measured daily for 7 days. On the last day interincisivus gap measured as the final gap. The difference between starting gap and the final gap calculated and have been analyzed by Variansi Analysis (Anava) to find the difference between the movement of controlling group and the group which have been given bisphosphonate risedronate per oral. To find the different effect between each risedronat dosage to the controlling used Post Hoc test. Research results showed that bisphosphonate risedronate per oral affected to avoid relapsing teeth to the rabbits after moved by the orthodontic utensils. There were found the meaningfull difference between controlling group and the group which were given bisphosphonate risedronate per oral (p<0,05). Minimum dosage bisphosphonate risedronate per oral to avoid relapsing rabbit teeth was 0,50 mg and it was more effective on 1 mg dosage.
