Gambaran darah dan histopatologik ikan mas (Cyprinus carpio) yang terinfeksi Koi Herpes Virus (KHV) secara alami

Tujuan penelitian ini adalah untuk mengetahui gambaran darah dan histopatologik ikan Mas yang terinfeksi secara alami oleh KHV. Sampel darah ikan Mas yang digunakan dalam penelitian ini berjumlah total 10 sampel, sebanyak 6 sampel berasal dari ikan Mas yang positif terdedah KHV, berdasarkan gejala klinis yang muncul di Waduk Cirata, Jawa Barat. Sebanyak 4 sampel lainnya diambil dari Cangkirngan yang tidak menunjukkan gejala klinis. Uji PCR IQ 2000 pada insang dilakukan di Laboratorium Balai Karantina Ikan Ngurah Rai, Denpasar, Bali. Penghitungan kadar sel eritrosit dan leukosit dengan metode Neubauer Counting Chamber, Packed Cell Volume (PVC) dengan metode mikrohematokrit, diferensial leukosit dengan metode pewarnaan Giemsa dilakukan di Laboratorium Patologi Klinik, Fakultas Kedokteran Hewan UGM. Pembuatan preparat histopatologik dengan teknik histologi dan pewarnaan Hematoksilin dan Eosin (HE) dilakukan di laboratorium Patologi Fakultas Kedokteran Hewan UGM. Data hasil analisa sampel darah secara laboratorium, dikaji secara statistik dengan t – test. Hasil pemeriksaan PCR menunjukkan positif terhadap KHV pada ikan Mas asal waduk Cirata (band 229 bp) dan negatif pada ikan Mas asal Cangkringan (band 665 bp). Hasil pemeriksaan darah menunjukkan adanya peningkatan (P<0,05) nilai leukosit (x Cangkringan = 6312,5 sel/μl; x Cirata = 19125 sel/μl) dan neutrofil ( x Cangkringan = 4065,75 sel/μl; x Cirata = 14302,33) pada ikan yang terinfeksi KHV. Organ ikan yang terinfeksi KHV mengalami keradangan pada insang, pankreas, hati, usus dan gonade; ginjal dan limpa mengalami kongesti. Benda inklusi intranuklear ditemukan pada epitel usus dan epitel lamela insang.

Research addressed to learn comprehensively the clinical pathological and histopathological change of common carp infected Koi Herpes Virus (KHV) naturally. Blood sample of common carp used in this research were sampled totally 10 samples, 6 samples originated common carp positively infected KHV. Based on clinical sign appear in Lake Cirata, West Java. While the others 4 samples were obtained from uninfected common carp regarded as group control. Examination by PCR test was conducted for both group samples. Erythrocyte and leucocyte counting by Neubauer Counting Chamber method, Packed Cell Volume (PVC) determined by microhematocrite method, differensial leucocyte determined using Giemsa staining method were worked in Laboratory of Clinical Pathology, Faculty of Veterinary Medicine, GMU. Histopathological preparation worked using histology technique and Hematoxilline and Eosin method at Laboratory of Pathology GMU. Haematological data resulted were then further analysed using t-test. Examination by PCR test result appeared positive to KHV on fish originated Lake Cirata (at band 229 bp) and negative for fish originated Cangkringan (at band 665 bp). Result of blood examination show us a significant change (p<0,05) of leukocyte (x Cangkringan = 6312,5 sel/μl; x Cirata = 19125 sel/μl) and neutrophile of KHV infected fish ( x Cangkringan = 4065,75 sel/μl; x Cirata = 14302,33) compared group Cangkringan. While. The fish tissue of KHV infected fish Inflamed such as : gill, pancreas, liver, intestine and gonad; while congestion were appeared within the spleen and ren microscopically. Inclusion body intranuclear was observed within intestine epithelial cell and lamella of the gill.
