Isolasi dan indentifikasi Escherichia coli O157H7 (Shiga-like toxin E. coli) pada sapi dan babi diare yang dilalulintaskan melalui karantina hewan Badas Sumbawa

2006 | Tesis | S2 Sain VeterinerPenelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Escherichia coli Serotipe O157 H7 (Shiga-like toxin E. coli) dari sapi dan babi diare yang dilalulintaskan melalui Karantina Hewan Badas- Sumbawa Sebanyak 100 sampel tinja yang diambil secara rektal masingmasing 50 ekor sapi dan 50 ekor babi diare yang akan dilalulintaskan. Sebelum pengambilan tinja dilakukan pencatatan terhadap umur dan asal ternak. Sampel tinja dimasukkan ke dalam media transport, disimpan dalam termos es suhu 5oC, selanjutnya dibawa ke Laboratorium Mikrobiologi Fakultas Kedokteran Hewan Universitas Gadjah Mada Yogyakarta. Jalan penelitian dilakukan dengan cara isolasi dan identifikasi berdasarkan sifat-sifat biokemisnya dan dilanjutkan dengan pemeriksaan serologis dengan uji aglutinasi tabung. Hasil menunjukkan berdasarkan sifat-sifat biokemia diperoleh 13 isolat (26 %) asal tinja sapi dan 19 isolat (38 %) asal tinja babi dan berdasarkan uji aglutinasi yang menunjukkan reaksi positif terhadap antiserum positif O157 H7 diperoleh isolat O157 H7 sebanyak 11 isolat (84,6 %) asal sapi dan 13 isolat (68,42 %) asal babi. Berdasarkan uji sensitivitas isolat E. coli O157 H7 terhadap antibiotika, menunjukkan semua isolat asal sapi (100 %) dan isolat asal babi (100 %) sensitif terhadap Enrofloxacin dan Tetracyclin, tetapi semua isolat sapi dan babi (100 %) resisten terhadap Streptomycin, Erythromycin dan Amoxycillin.
This research has purpose to isolate and identify Escherichia coli Serotipe O157 H7 ( Shiga-Like Toxin E. coli) from diarrhoea pig and bovine which shipping through Animal Quarantine of Badas Sumbawa A hundred faeces samples that was taken rectally of each 50 diarrhoea bovines and queues that would be shipped. Taking a note was done to livestock origin and age before taking the faeces. The samples were included into media transport, kept in ices thermos with temperature 5oC, and then the samples were brought to Microbiological Laboratory of Veterinary Faculty of Gadjah Mada Yogyakarta University. The research was conducted by isolation and identification based on the nature of biochemis and continued with inspection serologis with aglutinasi save test. The result that is based on the nature of biochemis showed it obtained 13 isolat ( 26 %) bovines faeces origin and 19 isolat ( 38 %) pigs faeces origin and based on the aglutinasi test that showed positive reaction to positive antiserum of O157 H7 obtained by isolat O157 H7 11 isolat ( 84,6 %)bovines origin and 13 isolat ( 68,42 %) pigs origin Based on sensitivity test of E. coli O157 H7 insolat to antibiotica, showing all isolat bovines origin ( 100 %) and isolat pigs origin ( 100 %) were sensitive to Enrofloxacin and Tetracyclin, but all isolat pigs and bovines ( 100 %) were resistant to Streptomycin, Erythromycin and Amoxycillin.
