Sitotoksisitas Tissue Conditioner terhadap biakan sel fibroblas gingiva manusia

2006 | Tesis | S2 Ilmu Kedokteran GigiTissue conditioner digunakan antara 2 hari sampai dengan 1 minggu. Kenyataannya pasien seringkali menggunakan tissue conditioner melebihi batas pemakaian. Tissue conditioner mengandung plasticizer yang dapat terlepas ke dalam saliva selama digunakan dan beberapa diantaranya diketahui bersifat toksik. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh tissue conditioner terhadap sitotoksisitas biakan sel fibroblas gingiva manusia. Penelitian dilakukan dengan menggunakan 78 sumur kultur sel fibroblas gingiva manusia. Kultur sel diberi paparan cuplikan hasil perendaman tissue conditioner dengan lama perendaman 3,7, 14, 21, dan 28 hari, kemudian diinkubasi selama 48 jam. Uji sitotoksisitas tissue conditioner dilakukan dengan uji MTT. Nilai OD dibaca dengan menggunakan ELISA plate reader pada panjang gelombang 550 nm. Perhitungan persentase kematian sel dilakukan berdasarkan nilai OD. Hasil penelitian didapatkan rerata persentase kematian sel pada perendaman 3 hari (18,086%), 7 hari (19,392%), 14 hari (45,874%), 21 hari (24,468%), dan 28 hari (18,640%), selanjutnya dianalisis menggunakan ANAVA satu jalur. Hasil analisis varian menunjukkan bahwa lama perendaman tissue conditioner berpengaruh bermakna terhadap persentase kematian sel (p<0,05). Persentase kematian sel meningkat sampai dengan lama perendaman 14 hari. Setelah perendaman 14 hari, persentase kematian sel kemudian mengalami penurunan.
Tissue conditioner is supposed to be used only between 2 days to 1 week. However, patients some times would use tissue conditioner more than the prescribed length of time. Tissue conditioner contains plasticizer that will leach out into the saliva during the usage. Moreover, some of the plasticizer is known to be toxic. This research aims to study the effect of tissue conditioner on citotoxicity of human fibroblast gingival cell culture. The research uses 78 wells of human fibroblast gingival cell culture. The cell culture is applied with soaked tissue conditioner with immersion times of 3, 7, 14, 21, and 28 days, then incubated for 48 hours. MTT test is used to detect the citotoxicity of the tissue conditioner and ELISA plate reader at 550 nm wave length to read the OD value. It relies on the OD value and average percentage of decayed cell at the immersion 3 days (18,086%), 7 days (19,392%), 14 days (45,874%), 21 days (24,468%), and 28 days (18,640%) are analyzed with one way ANOVA. The variant analysis results reveal that the length of tissue conditioner immersion time affects significantly to the percentage of fibroblast gingival cell decay (p<0.05). The percentage of cell decay increases up to 14 day immersion time. After that, the percentage decreases
