Pengaruh jumlah pengulangan Casting terhadap toksisitas Nickel Chromium Alloy pada biakan sel fibroblas gingiva

Laboratory process of recasting to dissolve cast alloy to perform a new restoration must effectively produce a biocompatible restoration without causing damage to tissue. The procedure itself must be safe and reduce any side-effect into a minimum. The research aims to study the effect of recasting frequency on nickel chromium alloy toxicity at human fibroblast gingival cell culture. The research used 96 wells of human fibroblast gingival cell culture. The cell culture was applied with soaked nickel chromium alloy without casting and 1, 2, 3, 4, 5 times recasting for 7-days and then incubated for 48 hours. MTT test was used to detect nickel chromium alloy toxicity and ELISA plate reader at a 550 nm wave length to read the OD value. It was relied on the OD value in the calculation of cell decay percentage and it was analyzed with one way ANOVA. The variant analysis results reveal that the number of recasting gives effect to the percentage of fibroblast gingival cell decay (p<0.05). The research concludes that the number of recasting affects fibroblast gingival cell toxicity, i.e., increasing the percentage of fibroblast cell decay. The highest percentage of cell decay (33 %) was obtained at 4 times recasting.
