Posisi Ruptur folikel dan kemampuan ovulasi Rattus norvegicus strain wistar setelah pemberian Pentagamavunon-0

Mekanisme kerja kurkumin sintetik dan PGV-0 pada sistem reproduksi sampai saat ini belum diketahui. PGV-0 merupakan analog kurkumin. Sampai saat ini belum ada penelitian tentang efek PGV-0 pada ruptur folikel sebagai pengatur fertilitas. Penelitian ini dilakukan untuk mengetahui posisi ruptur folikel dan kemampuan ovulasi ak ibat pemberian pentagamavunon-0 ( PGV -0 ), Sampel penelitian adalah folikel ovarium pada saat ovulasi dari tikus betina berumur 28 hari yang di induksi ovulasi dengan PMSG 5 IU ip, 48 jam kemudian disuntikka n hCG 5 IU i.p.Terdiri dari 6 kelompok yaitu: Kelompok tikus tanpa perlakuan (sebagai kontrol). Kelompok dua diberi CMC (pelarut), kelompok tiga diberi indometasin ED50, kelompok empat diberi PGV-0 ED50, Kelompok lima diberi PGV-0 2xED50 dan kelompok enam diberi PGV-0 4xED50. Zat perlakuan diberikan peroral pada umur 30 hari. Ovarium diambil pada umur 31 hari dan dibuat sedian irisan seri, diwarnai dengan Hematoxylin dan Eosin (HE) untuk dilakukan pangamatan posisi ruptur folikel. Analisa statistik menggunakan Kruskal Willis dan dilanjutkan dengan U – Mann Withney dengan tingkat signifikasi p<0,05. Tidak ada perbedaan posisi ruptur folikel antara kelompok control dan CMC (p>0,05).Perbedaan bermakna antara kelompok indometasin ED50 PGV-0 ED50, RGV-0 2X ED50 dan PGV-0 4x ED50 terhadap kelompok CMC (p<0,05) Tidak ada perbedaan bermakna antara kelompok EndometasinED50 terhadap kelompok PGV-0 berbagai dosis (p >0,05). Tidak ada perbedaan bermakna kemampuan ovulasi antara kelompok control dan klp CMC (p>0,05). Perbedaan bermakna ditemukan pada kelompok endometasin ED50, PGV-0 berbagai dosis terhadap klp CMC (p<0,05). Maka dapat disimpulkan bahwa PGV-0 dan indometasin menyebabkan ruptur folikel abnormal (terutama di basal) yang mempengaruhi kemampuan ovulasi.
Curcumin is a natural compound that has a lot of biological properties, including antifertility. The mechanism of curcumin synthesis and PGV-0 in reproductive system has not been known. PGV-0, an analog curcumin is an active ingredient of curcumin. Study on PGV-0 as fertility regulation on follicle rupture has not been carried out yet. This experiment was conducted to investigate the position of follicle rupture and ability of ovulation. The samples of this experiment were follicle of ovary at The time of ovulation obtained from ovaries induced immature rats, age 28 days old, by a sigle dose of 5 IU Pregnant mare serum gonadotropin (PMSG), and 48 hours later were injected with 5 IU of human chorionic gonadotropin (hCG). The immature rat were assign into 6 groups namely: the untreated rats (as control), the group treatment with CMC (solvent ), the group treatment with indometacin ED50, The groups treatment with PGV- ED50,the group treatment with PGV-0 2x ED50 and the last group treatment with PGV – 0 4x ED50. Treatment were given to rats orally at age 30 days as assign of groups. At the age of 31 days the rat were sacrificed and the ovaries were taken and examined microscopically after stained with Hematoxylin and Eosin (HE) to observe the position of follicle rupture and ovum in the tube of uterine. Statistic analysis using Kruskal Willis and followed by U – Mann Withney. A value of P<0,05 was considered to be significant. There was no significant difference between control group and CMC group (p> 0,05 ). There was no significanly difference between indometacin ED50 group and PGV-0 ED50 group (p> 0,05). There was no significantly difference ability of ovulation between control group and CMC group (p> 0,05). There was significantly difference between indometacin ED50 PGV-0 ED50, PGV – 0 2x ED50 and PGV-0 4 x ED 50 compare CMC group (p < 0,05), Conclution of study was: the position of follicle rupture were abnormal (more in basal) and decreased of ovulation rate in groups were treated with indometacin and PGV – 0.
