Drought stress is a major limiting factor that reduces sugarcane (Saccharum officinarum L.) productivity. The assembly of drought-tolerant sugarcane varieties can be achieved through genetic engineering by overexpressing the Dehydration-Responsive Element-Binding 2 (DREB2) gene. However, different sugarcane genotypes are hypothesized to exhibit varying responses to genetic transformation. Therefore, this study aimed to evaluate the differences in responsiveness between sugarcane genotypes VGM9901 and VGM0001 to the genetic transformation process and to obtain putative transformant plants. Genetic transformation was performed using Agrobacterium tumefaciens strain GV3101 harboring the pRI101ON binary plasmid carrying the DREB2 transgene. The in vitro stages included infection, co-cultivation, resting, three selection stages using a combination of cefotaxime and kanamycin antibiotics, and molecular confirmation via Polymerase Chain Reaction (PCR). Plantlet survival data were analyzed using the Independent Sample T-Test and Paired Sample T-Test at a 95% confidence level (?=0.05). The results showed that genotype VGM9901 consistently maintained a higher survival rate compared to VGM0001, indicating that VGM9901 is a more amenable genotype for genetic transformation. Paired T-Test analysis revealed that the most significant decline in explant population occurred during the transition from the Resting to the Selection 1 stage (p<0>DREB2 transformant plants were successfully obtained with a transformation efficiency ranging from 7.5% to 7.8%. Consequently, this study successfully established an optimal transformation protocol for sugarcane genetic transformation and provided critical insights regarding sugarcane genotypes responsive to DREB2 gene transformation.
