Kadar Glukosa Darah Tikus Putih (Rattus norvegicus L.) yang dibebani Glukosa setelah perlakuan ekstrak Daun Kluwih (Artocarpus altilis Park.) dan GABA (Gamma Amino Butyric Acid)

Tujuan penelitian ini adalah mengetahui kadar glukosa darah tikus putih yang dibebani glukosa dengan perlakuan ekstrak daun kluwih dan GABA serta mengetahui keberadaan/kadar GABA di dalam daun kluwih. Penelitian menggunakan 32 ekor tikus putih yang dibebani glukosa 4,5 g/kg BB per oral. Tiap sampel dibagi dalam delapan kelompok yaitu K1 kontrol normal, K2 dibebani glukosa tanpa perlakuan, K3 kelompok plasebo dengan akuades, K4 glibenklamid, K5 kelompok uji ekstrak etanol daun kluwih, K6 GABA dosis 0,0504 g/kg BB ip, K7 GABA dosis 0,1008 g/kg BB ip dan K8 kelompok uji ekstrak air daun kluwih. Sebelum perlakuan, kadar glukosa darah puasa diukur sebagai menit ke 0 dan pengukuran kembali diukur pada menit ke 60, 120 dan 180 setelah perlakuan. Pengukuran dilakukan pada hari ke-1, 7 perlakuan dan 7 hari setelah perlakuan dihentikan bersama dengan pengukuran berat badan. Hasil penelitian menunjukkan bahwa GABA murni mampu menurunkan kadar glukosa darah, ekstrak etanol lebih efektif menurunkan kadar glukosa darah dibanding ekstrak air daun kluwih, serta terdapat GABA dalam ekstrak etanol daun kluwih dengan kadar ± 0,01% dan ± 5,08% pada ekstrak air daun kluwih

The aims of this research are to know blood glucose level of rats Rattus norvegicus L. glucose loaded with treatment of breadnut leaf extract and GABA also GABA level in leaf of breadnut. The research was carried out which use 32 rats were oral glucose loaded 4,5 g/kg BW. Each samples was decided in eight groups; K1 normal control, K2 glucose group, K3 placebo group with akuades, K4 glibenclamide group, K5 etanol extract of breadnut leaf, K6 GABA 0,0504 g/kg BW ip, K7 GABA 0, 1008 g/kg BW ip and K8 water extract of breadnut leaf. Before treatment, fasting blood glucose data or called 0 minute were taken and 60, 120 and 180 minute after treatment, data were taken again. Do at 1st day, 7th days treatment and 7th days after treatment was stopped, together with measurement of body weight. The result show that pure GABA can decrease blood glucose level, etanol extract of breadnut leaf more effective decrease blood glucose level than water extract of breadfruit leaf and GABA level in etanol extract are ± 0,01% and ± 5,08% in the water extract of breadnut leaf.
