The identification and molecular characterisation of potential anti-cancer compounds from the ethyl acetate extract of the sponge Haliclona caerulea, sourced from the waters around Menjangan Kecil Island, Karimunjawa, have been carried out. This study aims to determine the toxicity of the extract or chromatographic fractions of the Haliclona caerulea sponge from the waters around Menjangan Kecil Island to Artemia salina using the Brine Shrimp Lethality Test (BSLT). Furthermore, the study also aimed to identify cytotoxic compounds from the ethyl acetate extract or chromatographic fractions of the Haliclona caerulea sponge. Extraction of the Haliclona caerulea sponge was carried out by maceration using methanol and dichloromethane as solvents; subsequently, the dichloromethane extract was partitioned with ethyl acetate and distilled water. The ethyl acetate extract was tested using the BSLT; the extract was then separated by column chromatography, and the resulting chromatographic fractions were also tested using the BSLT. The most toxic fraction, as determined by the BSLT, was identified using Liquid Chromatography-High Resolution Mass Spectrometry (LC-HRMS). The interactions of the identified compounds with the target protein CDK6 were analysed using molecular docking.
The isolation yielded a blackish-brown, paste-like ethyl acetate extract weighing 102.4 mg. The toxicity test results showed that the ethyl acetate extract of the Haliclona caerulea sponge was toxic to Artemia salina larvae, with a 50 per cent lethal concentration (LC??) of 97.247 ?g/mL. Separation by column chromatography yielded 11 combined fractions, with Fraction 3 being the most toxic. Based on LC-HRMS analysis and molecular docking, five potential compounds were identified from Fraction 3, namely (22E)-Cholesta-4,6,8(14),22-tetraen-3-one, Laingolide A, Conocenol D, Tanikolide and Pitholide B. All five compounds demonstrated interaction with amino acid residues at the CDK6 binding site. Laingolide A was selected as the most promising compound for further investigation based on its bond energy values and the similarity of its interaction patterns to those of palbociclib.
