Sorghum bagasse (Sorghum bicolor (L.) Moench) have potential as a feedstock for second-generation (G2) bioethanol production due to its abundant lignocellulosic content. However, the complex structure of lignocellulose hinders the efficiency of enzymatic hydrolysis, thus requiring a pretreatment stage. This study aimed to evaluate the effectiveness of microwave-assisted Deep Eutectic Solvent (DES) (ChCl–urea) in the delignification process, to determine the optimum conditions for the combination of biomass ratio, processing time, and DES concentration using Response Surface Methodology (RSM) with a Box-Behnken Design (BBD), and measure reducing sugar yields following enzymatic hydrolysis. The independent variables tested comprised the biomass-to-DES solution ratio (8%, 10% and 12% w/w), processing time (5, 10 and 15 minutes), and DES concentration (1%, 1.5% and 2% w/v), designed across 15 experimental runs. The results of the study showed that DES pretreatment successfully increased the cellulose content from 37.95% to 45.32%, reduced the lignin content from 19.20% to 14.41%, and reduced the hemicellulose content from 29.81% to 24.60%. Optimum conditions were achieved at a biomass ratio of 10.7%, a processing time of 15 minutes, and a DES concentration of 1.9%, with a predicted cellulose content of 48.16% and an actual yield of 45.32%. Analysis of reducing sugars using the Nelson-Somogyi method yielded 8.77 mg/mL (8.77 g/L), with a yield coefficient of 0.1754 g/g and a cellulose conversion efficiency of 34.84%. The microwave-assisted DES ChCl-urea pretreatment method proved effective in enhancing cellulose accessibility for reducing sugar production as a bioethanol feedstock.
