Spirodela polyrhiza is a protein-rich aquatic plant with high potential as an alternative poultry feed; however, its utilization is still hindered by its complex fiber (lignocellulose) content and antinutritional factors. This study aimed to test and evaluate the effects of fermentation using a single isolate of Lactiplantibacillus plantarum FNCC 0026 and a bacterial consortium including Bacillus subtilis FNCC 0059 on changes in the proximate nutritional quality, amino acid profile, and micromorphological characteristics of S. polyrhiza. This study included the following fermentation treatments: no inoculation/control (P0), inoculation with a single isolate of L. plantarum (P1), and inoculation with a bacterial consortium (P2). Feed quality was evaluated through proximate analysis, analysis of the substrate’s surface microstructure using Scanning Electron Microscopy (SEM), and confirmation of microbial taxonomic identity via 16S rRNA gene phylogenetic tree analysis. The results of the proximate analysis showed that fermentation was able to increase the Crude Protein (CP) content from 23.75% (fresh fronds) to 27,47% (P1) and 24,30% (P2) and reduced the Crude Fiber (CF) content from 16.13% to 9,59% (P1) and 12,06% (P2). SEM observations at 5.000× magnification confirmed the presence of massive bacterial cell colonies on the surface of the duckweed substrate matrix, while phylogenetic analysis based on 16S rRNA gene sequences was used to examine the genetic relationships between the bacterial sequences detected in the fermentation samples and the reference sequences. Overall, the fermentation of Spirodela polyrhiza using bacterial inoculants proved in enhancing nutritional value, breaking down fiber, and enriching the amino acid profile, thereby demonstrating its potential as a high-protein feed.
