Duckweed flour shows potential as an alternative poultry feed ingredient due to its high protein content and diverse amino acid profile; however, its utilization is constrained by a high crude fiber content. Bacillus subtilis-based fermentation has the potential to improve its nutritional quality, yet the application of B. subtilis on duckweed flour remains sparsely explored. This study aimed to evaluate the capacity of B. subtilis FNCC 0059 to ferment duckweed flour. Fermentation was conducted on sterile duckweed flour with the addition of molasses and bacterial inoculants (P0: control, P1: +B. subtilis FNCC 0059, and P2: +B. subtilis FNCC 0059 and Lactiplantibacillus plantarum FNCC 0026) for 96 hours. Observed parameters included pH value, proximate composition, surface characteristic analysis via Scanning Electron Microscopy (SEM), and 16S rRNA molecular identification. The results demonstrated that the pH value decreased from 6.5 to 6.0 across all treatments. Crude fiber content declined from 16.13% to 10.66% (P0), 9.46% (P1), and 12.06% (P2), while crude protein increased from 23.75% to 27.55% (P0), 27.10% (P1), and 24.30% (P2). SEM observation at 5,000x magnification confirmed the presence of cell colonies on the surface of the duckweed substrate matrix. However, changes in proximate composition under the bacterial inoculation treatments yielded values comparable to the control without inoculants, and 16S rRNA gene sequencing isolated from the sample was identified as Priestia megaterium. Based on this study, the addition of B. subtilis FNCC 0059 was not proven to exert a significant effect on the fermentation process of duckweed flour compared to spontaneous fermentation.
