Sunflower (Helianthus annuus L.) leaf biomass is generally underutilized because leaves are not harvested as the primary product, whether the crop is cultivated for seed production or as an ornamental landscape plant. Sunflower leaves contain allelopathic secondary metabolites and therefore have potential as a raw material for bioherbicide production. Arbuscular mycorrhizal fungal inoculation may enhance plant growth and primary yield while promoting the accumulation of secondary metabolites in the leaves. This study aimed to evaluate the responses of eight sunflower genotypes to arbuscular mycorrhizal fungal inoculation based on root colonization, agronomic traits, secondary metabolites, and the expression of genes involved in the phenylpropanoid pathway, as well as to select a genotype with potential as a source of leaf extract for bioherbicide production without compromising the crop’s primary yield. Genotypic responses were evaluated using a factorial randomized complete block design, while genotype selection integrated performance under mycorrhizal conditions and relative responses to mycorrhizal inoculation in terms of the crop’s primary functions and bioherbicide-related traits. Leaf extracts from the selected genotype were subsequently evaluated using a factorial completely randomized design, with extract source and concentration as factors, to determine their inhibitory effects on germination and early growth of purple nutsedge (Cyperus rotundus L.) as the target weed and black soybean (Glycine max L.) as the cultivated crop. The results showed that all genotypes were able to establish symbiosis with arbuscular mycorrhizal fungi. Mycorrhizal inoculation generally increased root colonization relative to the control, with descriptive relative increases ranging from 167.6% to 275.8%. It also generally improved several vegetative traits and plant biomass, whereas responses in generative traits and secondary metabolite contents were genotype-dependent. The expression of genes involved in the phenylpropanoid pathway differed among genotypes but was not significantly affected by mycorrhizal inoculation. The integration of performance under mycorrhizal conditions and relative responses to mycorrhizal inoculation identified Ha 1 as the selected genotype because it exhibited increases in seed yield components as well as leaf biomass, total phenolic content, and free ferulic acid content. Ha 1 was subsequently used as the source of leaf extracts to evaluate their inhibitory activity against the target weed and cultivated crop. Ha 1 leaf extracts inhibited the growth of purple nutsedge more strongly than that of black soybean, particularly during the early stage of tuber sprouting, with inhibition increasing as extract concentration increased. Extracts derived from control and mycorrhizal plants did not differ significantly in their effects, although those from mycorrhizal plants tended to exhibit stronger inhibitory activity. These findings indicate that Ha 1 leaf biomass has potential as a raw material for bioherbicide production to suppress the early growth of purple nutsedge; however, its selectivity toward cultivated crops requires further evaluation.
