Aktivitas antikanker dan prediksi mekanisme kerja turunan kurkumin mengandung boron (CCB-4) terhadap sel kanker payudara triple negative (MDA-MB-231)

Kanker payudara masih menjadi penyebab kematian tertinggi pada wanita. Salah satu subtipe kanker kanker payudara adalah Triple Negative Breast Cancer (TNBC) yang bersifat lebih agresif dengan prognosis yang buruk dan pilihan terapi yang terbatas. Kemoterapi konvensional masih menjadi terapi utama, tetapi toksisitasnya terhadap sel normal mendorong pengembangan kandidat senyawa antikanker yang lebih efektif dan selektif. Chemoprevention Curcumin Boron-4 (CCB-4) merupakan turunan kurkumin yang dimodifikasi dengan penambahan gugus boron untuk meningkatkan aktivitas biologis dan selektivitas antikanker. Penelitian ini bertujuan mengevaluasi aktivitas antikanker CCB-4 terhadap sel TNBC, yaitu MDA-MB-231, melalui pendekatan in vitro dan in silico. Aktivitas sitotoksik dan selektivitas dievaluasi menggunakan uji MTT. Kemampuan CCB-4 dalam menginduksi apoptosis dan senescence dianalisis masing-masing menggunakan DNA fragmentation assay dan pewarnaan  SA-?-gal. Aktivitas penghambatan migrasi CCB-4 dievaluasi menggunakan scratch wound healing assay. Selain itu, dilakukan juga prediksi mekanisme kerja CCB-4 melalui identifikasi target protein dan molecular docking terhadap protein-protein yang berperan dalam progresi TNBC.
Hasil penelitian menunjukkan bahwa CCB-4 memiliki aktivitas sitotoksik yang kuat terhadap sel MDA-MB-231 dengan IC50 sebesar 2,1 ± 0,2 µM dan doksorubisin sebesar 3,8 ± 1,0 µM. Senyawa CCB-4 bersifat lebih selektif terhadap sel kanker, ditunjukkan oleh nilai SI ? 2, yaitu sebesar 4,14 dibandingkan dengan doksorubisin sebesar 1,44. Berdasarkan uji kualitatif fragmentasi DNA, CCB-4 berpotensi memiliki kemampuan menginduksi apoptosis yang ditunjukkan oleh adanya pola pita fragmen DNA melalui elektroforesis. CCB-4 juga berpotensi menginduksi senescence, yang ditandai dengan meningkatnya sel yang positif SA-?-gal. Perlakuan CCB-4 terhadap MDA-MB-231 pada konsentrasi IC50 menunjukkan penghambatan migrasi. Pada inkubasi 24 jam, konsentrasi IC50 CCB-4 menurunkan  persentase penutupan luka (%wound closure) sebesar  87,69 ± 1,62% (p?0,05) dibandingkan dengan kontrol sebesar 93,24 ± 0,55%. Analisis bioinformatika memprediksi AKT1 dan PTGS2 sebagai protein target utama CCB-4 terhadap TNBC. Protein tersebut berperan dalam  jalur persinyalan PI3K/AKT1 yang mengatur proliferasi sel, survival sel, dan regulasi inflamasi. CCB-4 diprediksi memiliki pengikatan pada kedua protein tersebut dengan energi pengikatan sebesar -8,83 kkal/mol pada AKT1 dan -6,32 kkal/mol. Berdasarkan hasil penelitian, CCB-4 menunjukkan potensi sebagai kandidat antikanker terhadap TNBC karena memiliki aktivitas sitotoksik yang kuat dan lebih selektif terhadap sel kanker dibandingkan dengan doksorubisin, disertai dengan adanya induksi apoptosis dan senescence, serta menghambat migrasi. Analisis bioinformatika dan molecular docking juga mendukung bahwa aktivitas tersebut kemungkinan melibatkan modulasi protein AKT1 dan PTGS2 yang berperan dalam progresi TNBC. 

Breast cancer remains the leading cause of cancer-related mortality among women globally. Triple-negative breast cancer (TNBC) represents an aggressive subtype with a poor prognosis and limited therapeutic options, rendering conventional chemotherapy the primary treatment. Chemotherapy, however, frequently causes toxicity to normal cells and leads to adverse side effects. Therefore, the urgent need for more effective and selective anticancer agents persists. Chemoprevention Curcumin Boron-4 (CCB-4), a boron-containing curcumin derivative, was developed to enhance biological activity and anticancer selectivity. The present study evaluated the anticancer activity of CCB-4 against the TNBC cell line MDA-MB-231 using both in vitro and in silico methodologies. Cytotoxicity and selectivity were assessed via MTT assay. Apoptosis and senescence induction by CCB-4 were examined using DNA fragmentation and SA-?-gal staining, respectively. Anti-migratory effects were determined through the scratch wound healing assay. Additionally, the molecular mechanism of CCB-4 was investigated by protein target identification and molecular docking analyses involving proteins implicated in TNBC progression.
The results demonstrated that CCB-4 exhibited potent cytotoxic activity against MDA-MB-231 cells, with an IC50 of 2,1 ±  0,2 µM, compared to 3,2 ± 1,0 µM for doxorubicin. CCB-4 also exhibited greater selectivity for cancer cells, with a selectivity index (SI) of 4,14 and 1,44 for doxorubicin. Qualitative DNA fragmentation analysis revealed that CCB-4 induced apoptosis, as indicated by the characteristic DNA laddering pattern observed after agarose gel electrophoresis. CCB-4 further induced cellular senescence, as shown by an increased proportion of SA-?-gal-positive cells. At the IC50 concentration, CCB-4 inhibited cell migration, resulting in a wound closure percentage of 87,69 ± 1,62 ?ter 24 hours of incubation, compared to 93,24 ± 0,55% in the untreated control (p? 0,05). Bioinformatics analysis identified AKT1 and PTGS2 as primary predicted molecular targets of CCB-4 in TNBC. These proteins are in the PI3K/AKT signaling pathway, which regulates cell proliferation, cell survival, and inflammatory responses. Molecular docking analysis predicted favorable binding affinities of CCB-4 to AKT1 and PTGS2, with binding energies of -8,83 kcal/mol and -6,32 kcal/mol, respectively. Overall, these findings suggest that CCB-4 is a promising anticancer candidate for TNBC, exhibiting potent cytotoxicity, greater selectivity than doxorubicin, the ability to induce apoptosis and senescence, and inhibition of cell migration. Bioinformatics and molecular  docking analyses further indicate that these effects may involve modulation of AKT1 and PTGS2, proteins implicated in TNBC progression.
