PENGARUH EKSTRAK DAUN MENGKUDU (Morinda citrifolia L.) TERHADAP HIDROFOBISITAS BAKTERI STREPTOCOCCUS VIRIDANS (Kajian secara in vitro)

Perlekatannya pada pelikel gigi berperan penting dalam pembentukan plak gigi. Perlekatan awal bakteri Streptococcus viridans pada pelikel gigi sangat dipengaruhi oleh hidrofobisitas bakteri tersebut. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh ekstrak daun mengkudu (Morinda citrifolia L.) terhadap hidrofobisitas bakteri Streptococcus viridans. Daun Morinda citrifolia L. diekstraksi secara maserasi hingga mencapai konsentrasi 1%, 2%, dan 4%. Bakteri Streptococcus viridans yang diisolasi dari plak gigi manusia dipaparkan ekstrak Morinda citrifolia L. dengan berbagai konsentrasi, akuades sebagai kontrol negatif, dan Klorheksidin glukonat 0,12% sebagai kontrol positif. Semua kelompok perlakuan ditambah dengan heksadekana, yaitu suatu larutan hidrokarbon yang memiliki pelekatan pada rongga mulut. Pengaruh ekstrak Morinda citrifolia L. terhadap hidrofobisitas bakteri Streptococcus viridans diketahui dengan mengukur nilai densitas optik pada spektrofotometer. Panjang gelombang yang digunakan pada spektrofotometer adalah 600 nm. Hasil uji One way Anava menunjukkan adanya perbedaan nilai indeks hidrofobisitas bakteri Streptococcus viridans secara bermakna antara kelompok uji dan kelompok kontrol (p<0,05). Indeks hidrofobisitas terkecil bakteri Streptococcus viridans terlihat pada pemaparan ekstrak daun Morinda citrifolia L. konsentrasi 4%. Kesimpulan dari penelitian ini yaitu ekstrak daun mengkudu (Morinda citrifolia L.) berpengaruh menurunkan hidrofobisitas bakteri Streptococcus viridans.
Species of Streptococcus viridans group have been known as commensal bacteria that dominate the human oral cavity. Its attachment to the acquired pellicle play an important role in the formation of dental plaque. Initial attachment of Streptococcus viridans in acquired pellicle was strongly influenced by the hydrophobicity of the bacteria. The purpose of this study was to determine the effect of noni leaf extract (Morinda citrifolia L.) against the hydrophobicity of Streptococcus viridans. Leaves of Morinda citrifolia L. were extracted by maceration to achieve a concentration of 1%, 2%, and 4%. Streptococcus viridans bacteria that was isolated from human dental plaque exposed by Morinda citrifolia L. extracts with various concentrations as a treatment group, distilled water as a negative control, and Chlorhexidine gluconate 0,12% as a positive control. All treatment groups were added with hexadecane, which is a hydrocarbon solution representing acquired pellicle in the oral cavity. Effect of extract of Morinda citrifolia L. to the hydrophobicity of Streptococcus viridans was known by measuring the optical density values on the spectrophotometer. Wavelength that used in spectrophotometer was 600 nm. One way Anova test results indicated a significant difference in the hydrophobicity index values of the bacterium Streptococcus viridans between the treatment group and control group (p<0.05). Smallest hydrophobicity index of Streptococcus viridans was seen in the exposure of Morinda citrifolia L. leaf extract concentration of 4%. The conclusion from this study that the leaf extract of noni (Morinda citrifolia L.) affects to reduce the hydrophobicity of Streptococcus viridans.
