Aktivitas Antibakteri Ekstrak Tunggal dan Kombinasi Daun Kumis Kucing dan Daun Tempuyung terhadap Bakteri Pseudomonas aeruginosa secara in vitro

Latar Belakang : Batu ginjal dapat memicu Infeksi Saluran Kemih (ISK), karena sumbatan akibat batu menciptakan kondisi yang mendukung pertumbuhan bakteri seperti P. aeruginosa. Daun tempuyung dikenal mampu meluruhkan batu ginjal, sehingga membantu mencegah terjadinya ISK, sementara daun kumis kucing mengandung senyawa bioaktif yang berpotensi langsung menghambat pertumbuhan bakteri. Kombinasi kedua ekstrak ini diharapkan dapat menjadi alternatif terapi yang lebih efektif pada ISK akibat batu ginjal.

Tujuan : Penelitian ini bertujuan untuk mengkaji aktivitas antibakteri ekstrak tunggal maupun kombinasi daun kumis kucing dan daun tempuyung dengan empat perbandingan kadar berbeda terhadap pertumbuhan bakteri P. aeruginosa menggunakan metode difusi agar, serta mengetahui profil metabolit dari kedua ekstrak tersebut melalui analisis KLT.

Metode : Pembuatan ekstrak tunggal maupun kombinasi menggunakan metode maserasi dengan pelarut etanol 70%. Uji Fitokimia untuk mengetahui profil golongan senyawa dalam kedua ekstrak menggunakan metode KLT. Pengujian Antibakteri menggunakan metode difusi yaitu menggunakan kertas cakram pada media Muller Hinton (MHA).

Hasil : Uji difusi cakram menunjukkan ekstrak tunggal maupun kombinasi daun kumis kucing dan tempuyung hanya menghasilkan zona hambat kecil terhadap P. aeruginosa (0,75–1,70 mm), dengan kombinasi 100% (Tempuyung : Kumis Kucing) memberikan hambatan terbesar (1,70 ± 1,57 mm), namun masih jauh lebih rendah dibanding kontrol positif kloramfenikol (9,07 ± 1,81 mm), tanpa perbedaan signifikan antar perlakuan. Analisis KLT menunjukkan bahwa pada ekstrak kumis kucing, terdeteksi tujuh bercak pada UV 366 nm dengan rentang Rf 0,24–0,96, sedangkan pada UV 254 nm hanya tampak satu bercak dominan pada Rf = 0,96. Pada ekstrak tempuyung, baik pada UV 366 nm maupun UV 254 nm terdeteksi tujuh bercak dengan rentang Rf 0,15–0,95.

Kesimpulan : Ekstrak tunggal dan kombinasi daun kumis kucing serta tempuyung mampu menghambat pertumbuhan P. aeruginosa meskipun dengan efek lemah, profil KLT menunjukkan keberadaan metabolit sekunder yang berpotensi sebagai agen antibakteri untuk penelitian lanjutan.

Background: Kidney stones can trigger urinary tract infections (UTI), as obstruction caused by the stones creates favorable conditions for bacterial growth such as P. aeruginosa. Tempuyung leaves are known to help dissolve kidney stones, thus preventing UTI, while kumis kucing contains bioactive compounds that can directly inhibit bacterial growth. The combination of these extracts is expected to provide a more effective alternative therapy for UTI caused by kidney stones.

Objective: This study aims to evaluate the antibacterial activity of single and combined extracts of kumis kucing and tempuyung with four different concentration ratios against the growth of P. aeruginosa using the agar diffusion method, as well as to determine the metabolite profile of both extracts throughTLC analysis.

Methods: The single and combined extracts were prepared using the maceration method with 70% ethanol as the solvent. Phytochemical screening was carried out to identify the classes of compounds in both extracts using TLC. The antibacterial activity was tested using the diffusion method with paper discs on Muller Hinton Agar (MHA) medium.

Results : The disc diffusion assay showed that both single and combined extract of kumis kucing and tempuyung produced only small inhibition zones against P. aeruginosa (0.75–1.70 mm). The highest inhibition was observed in the 100% combination of Tempuyung : Kumis Kucing (1.70 ± 1.57 mm), which remained markedly lower than the positive control, chloramphenicol (9.07 ± 1.81 mm), with no significant differences among treatments. TLC analysis revealed that the kumis kucing extract exhibited seven spots under UV 366 nm with Rf values ranging from 0.24 to 0.96, whereas only one dominant spot (Rf = 0.96) was visible under UV 254 nm. In contrast, the tempuyung extract displayed seven spots under both UV 366 nm and UV 254 nm, with Rf values ranging from 0.15 to 0.95.

Conclusion : Single and combined extracts of kumis kucing and tempuyung were able to inhibit P. aeruginosa growth, although the effect was weak. TLC profile showed the presence of secondary metabolites that may contribute to antibacterial potential.
