Karakterisasi Patogen dan Profil Senyawa Metabolit Terkait Patosistem Kelapa Sawit-Ganoderma

Kelapa sawit (Elaeis guineensis Jacq.) merupakan komoditas strategis yang berperan penting dalam perekonomian Indonesia. Namun, produktivitasnya menghadapi ancaman serius dari penyakit busuk pangkal batang (BPB) yang disebabkan oleh Ganoderma. Penyakit ini sulit dikendalikan dan dapat menyerang tanaman sejak fase pembibitan hingga tanaman menghasilkan. Keberagaman spesies dan tingkat virulensi Ganoderma menuntut identifikasi patogen yang akurat serta pemahaman terhadap respons tanaman setelah infeksi. Penelitian ini bertujuan untuk (1) mengidentifikasi isolat Ganoderma dari Sumatra sebagai penyebab BPB secara morfologi dan molekuler serta mengkarakterisasinya secara biokimia dan berdasarkan profil genetik menggunakan rep-PCR; (2) mengetahui tingkat virulensi isolat Ganoderma asal Sumatra pada bibit kelapa sawit; dan (3) mengidentifikasi senyawa metabolit yang terkait dengan patosistem kelapa sawit–Ganoderma. Penelitian dilaksanakan dalam tiga tahap, yaitu identifikasi dan karakterisasi isolat Ganoderma, pengujian virulensi isolat pada bibit kelapa sawit, serta analisis profil metabolit yang terkait dengan patosistem kelapa sawit–Ganoderma. Identifikasi molekuler secara multilokus terhadap 41 isolat dari tujuh provinsi di Sumatra menunjukkan bahwa isolat-isolat tersebut termasuk dalam dua spesies, yaitu Ganoderma boninense dan G. ellipsoideum. Kedua spesies menunjukkan variasi karakter biokimia dan profil genetik berdasarkan rep-PCR. Dari 14 isolat G. boninense, empat menunjukkan virulensi tinggi (GLLS-N002A, GSUS-003, GRK-K005, dan GRK-K001). Kedua isolat G. ellipsoideum memiliki virulensi sangat rendah. Analisis metabolomik menunjukkan perbedaan profil metabolit daun pada varietas PPKS 540 NG dan PPKS 239 yang diinokulasi dengan dua isolat G. boninense berbeda virulensi (GLLS-N002A dan GRIHU-S002). Allantoin berpotensi menjadi kandidat biomarker tentatif keberadaan infeksi pada fase awal karena muncul secara konsisten, sedangkan acetoacetate berpotensi menjadi indikator respons pertahanan pada PPKS 540 NG, serta betaine dan TMAO berpotensi menjadi indikator stres fisiologis dan perkembangan penyakit pada PPKS 239. Hasil penelitian memberikan informasi mengenai keragaman Ganoderma, tingkat virulensi, serta perubahan metabolit yang terkait dengan patosistem kelapa sawit–Ganoderma sebagai dasar pengembangan strategi deteksi dan pengelolaan penyakit BPB.
Oil palm (Elaeis guineensis Jacq.) plays a role in the Indonesian economy. However, its productivity is threatened by basal stem rot (BSR) caused by Ganoderma. The disease is hard to control, affecting plants from nursery to productive stage. The diversity of Ganoderma species and their virulence levels necessitate pathogen identification and an understanding of infection responses. This study aimed to (1) identify Ganoderma isolates from Sumatra as causal agents of BSR based on morphological and molecular characteristics and characterize them biochemically and genetically using rep-PCR; (2) determine the virulence levels of Ganoderma isolates on oil palm seedlings; and (3) identify metabolites associated with the oil palm–Ganoderma pathosystem. The study included three stages: identification and characterization of Ganoderma isolates, evaluation of isolate virulence, and metabolite profiling. Multilocus molecular identification of 41 isolates from seven provinces in Sumatra showed that they belonged to two species, Ganoderma boninense and G. ellipsoideum. The species exhibited variation in biochemical characteristics and genetic profiles based on rep-PCR. Among 14 G. boninense isolates, four showed high virulence (GLLS-N002A, GSUS-003, GRK-K005, and GRK-K001), whereas G. ellipsoideum isolates exhibited very low virulence. Metabolomic analysis revealed differences in leaf metabolite profiles between PPKS 540 NG and PPKS 239 inoculated with GLLS-N002A and GRIHU-S002. Allantoin showed potential as a tentative biomarker of early infection, acetoacetate as an indicator of defense response in PPKS 540 NG, and betaine and TMAO as indicators of physiological stress and disease progression in PPKS 239. The findings provide a basis for BSR detection and management strategies.
