berbagai sampel lingkungan domestik, kemudian profil resistensi antibiotik (ampisilin,
Antibiotic
resistance in Escherichia coli has become a major concern, prompting the
development of alternative strategies for bacterial control, one of which is
the use of bacteriophages. This study aimed to isolate and identify E. coli
isolates from domestic environmental samples; to examine and characterize
antibiotic-resistant E. coli isolates through antibiotic susceptibility
profiling; to evaluate the lytic activity of single and cocktail bacteriophages
against E. coli isolates from domestic environments; and to compare the
lytic activity of single and cocktail bacteriophages against
antibiotic-resistant E. coli isolates. E. coli isolation and
identification were performed using various domestic environmental samples,
followed by antibiotic susceptibility testing against ampicillin,
ciprofloxacin, and tetracycline using the disk diffusion method. Bacteriophage
lytic activity was assessed through host range analysis (spot test) and comparative
evaluation of single and cocktail phage lytic activity using domestic
wastewater isolates as host culture indicators. The results showed that several
E. coli isolates exhibited antibiotic resistance, and both single and
cocktail bacteriophages were able to lyse the target isolates with varying
degrees of effectiveness. In contrast, isolates ECKS, ECAM, and ECKN showed no
lysis under any treatment (-), indicating resistance to the bacteriophages
used. In isolate ECSK, phages SAV and SCV produced weak lysis (+), whereas
phages SBV, SEV, and the phage cocktail resulted in partial lysis (++).
Overall, the bacteriophage cocktail exhibited a broader lytic activity spectrum
than individual bacteriophages. Therefore, bacteriophages have potential to be
developed as biocontrol agents against antibiotic-resistant E. coli
originating from domestic environments.
