Background: Excessive physical activity leads to muscle fatigue and injury. Macrophages undergo phagocytosis and produce pro-inflammatory cytokines, one of which is TNF-alpha. TNF-alpha exacerbates inflammation by increasing the transcription of pro-inflammatory cytokines in the nucleus via the NF-kappa B pathway. Consequently, prostaglandin levels rise, resulting in the clinical manifestations of inflammation. Herbal plants such as temulawak (Curcuma xanthorrhiza), turmeric (Curcuma longa) and meniran (Phyllanthus niruri) contain active compounds with the potential to reduce the expression of TNF-alpha and PTGES.
Objective: This study aimed to determine the protective effects of a polyherbal formulation C. xanthorrhiza, C. longa and P. niruri on TNF-alpha and PTGES mRNA expression in mice subjected to exhaustive swimming.
Methods: TNF-alpha and PTGES mRNA expression in mice gastrocnemius muscles obtained from Sholiha (2025) were analyzed using the qPCR method. Sholiha utilized five groups of mice (n=5 per group): a normal group (distilled water) and four exhaustive swimming groupscomprising a negative control (distilled water) and three treatment groups receiving doses of 78 mg/kg, 156 mg/kg, and 312 mg/kg. The polyherbal extract was administered orally for 28 days, with exhaustive swimming sessions conducted on days 26, 27, and 28, 30 minutes after extract administration. Differences in mean mRNA expression among groups were statistically analyzed using one-way ANOVA followed by a post-hoc LSD test. A p-value of <0>
Results: TNF-alpha mRNA expression in the normal group was lower than in the negative control group; furthermore, higher doses of the polyherbal extract resulted in higher TNF-alpha mRNA expression compared to the negative control. The highest TNF-alpha mRNA [removed]17.61 ± 4.592) was observed in the dose 3 group, a result that was statistically significant (p<0>
Conclusion:TNF-alpha mRNA expression in mice administered the polyherbal extract at dose 3 was higher compared to the control group that did not receive the polyherbal extract. PTGES mRNA expression in mice administered the polyherbal extract at doses 2 and 3 was lower compared to the control group that did not receive the polyherbal extract.
