2026 | Tesis | S2 BioteknologiArachnis flos-aeris merupakan anggrek tropis dengan nilai estetika dan ekonomi tinggi, namun pertumbuhan batang yang relatif tinggi membatasi efisiensi budidaya, transportasi, dan pemanfaatannya sebagai tanaman hias. Penyuntingan genom menggunakan sistem CRISPR/Cas9 yang menargetkan gen GAI (Gibberellic Acid Insensitive), salah satu gen penyandi protein DELLA sebagai regulator negatif jalur pensinyalan giberelin, merupakan pendekatan yang potensial dengan tujuan untuk menghasilkan A. flos-aeris berfenotip kerdil dengan tetap mempertahankan estetika dan ukuran bunganya. Penelitian ini meliputi optimasi perkecambahan biji secara in vitro, isolasi dan karakterisasi gen GAI, desain dan konstruksi sgRNA ke dalam vektor pRGEB32, transformasi plasmid ke Agrobacterium tumefaciens strain GV3101, serta transformasi genetik pada protokorm melalui metode perendaman. Keberhasilan transformasi dikonfirmasi melalui deteksi gen Cas9 menggunakan polymerase chain reaction (PCR), sedangkan penyuntingan gen GAI diverifikasi melalui analisis sekuens DNA. Hasil penelitian menunjukkan bahwa media NP yang diperkaya ekstrak tomat 100 g/L merupakan media paling efektif untuk mendukung perkembangan embrio hingga fase 5. Penambahan asetosiringon 100 µM menghasilkan efisiensi transformasi tertinggi sebesar 12,59%. Analisis molekuler mengonfirmasi keberhasilan integrasi konstruksi T-DNA UBI::CRISPR/Cas9::U3::GAI ke dalam genom A. flos-aeris serta mengidentifikasi mutasi berupa substitusi dan insersi pada daerah target gen GAI di domain DELLA. Tanaman transforman menunjukkan perubahan morfologi berupa penurunan tinggi dan lebar tanaman serta pertumbuhan yang lebih kompak, disertai perubahan anatomi berupa penurunan ukuran sel protomeristem dibandingkan wild type. Hasil penelitian ini memberikan dasar ilmiah bagi pengembangan varietas anggrek mini A. flos-aeris melalui penerapan penyuntingan genom menggunakan sistem CRISPR/Cas9, sehingga berpotensi meningkatkan nilai estetika, nilai komersial, serta efisiensi pemanfaatannya sebagai tanaman hias pada ruang yang terbatas.
Arachnis flos-aeris is a tropical orchid with high ornamental and economic value; but its relatively tall stem growth limits cultivation efficiency, transportation, and utilization as an ornamental plant in limited spaces. Genome editing using the CRISPR/Cas9 system targeting the GAI (Gibberellic Acid Insensitive) gene, which encodes the DELLA protein as a key negative regulator of the gibberellin signaling pathway, represents a promising approach with the aim of generating dwarf A. flos-aeris plants while maintaining their ornamental value and flower size. This study involved in vitro seed germination optimization, isolation and characterization of the GAI gene, sgRNA design and construction into the pRGEB32 vector, plasmid transformation into Agrobacterium tumefaciens strain GV3101, and genetic transformation of orchid protocorms using the immersion method. Transformation was confirmed by detecting the Cas9 gene using PCR, while genome editing was verified through DNA sequence analysis. The results showed that NP medium supplemented with 100 g/L tomato extract was the most effective medium for promoting embryo development to protocorm phase 5. Supplementation with 100 ?M acetosyringone produced the highest transformation efficiency of 12,59%. Molecular analysis confirmed the successful integration of the UBI::CRISPR/Cas9::U3::GAI T-DNA construct into the A. flos-aeris genome and identified substitution and insertion mutations within the target region of the GAI gene in the DELLA domain. The transformed plants exhibited morphological changes characterized by reduced plant height and width and a more compact growth habit, accompanied by anatomical changes including smaller protomeristem cell size compared with the wild-type plants. These findings provide a scientific basis for developing A. flos-aeris mini orchid through CRISPR/Cas9-mediated genome editing, offering potential to enhance ornamental value, commercial value, and cultivation efficiency in limited spaces.
