This study was motivated by the limited intrinsic contrast of biological tissues in Photoacoustic Imaging (PAI), which necessitates the development of optical contrast agents capable of enhancing signal contrast between target tissues and the surrounding background. One promising candidate is Carbon Dots (CDs), which possess favorable optical and thermal properties, nanoscale dimensions, biocompatibility, and absorption potential in the near-infrared (NIR) region. This study aimed to investigate the effect of CDs-based contrast agents on improving the imaging quality of salivary glands using a modulated laser-based PAI system. The research involved the synthesis of CDs from soursop leaves using a hydrothermal method with gradual heating up to 150 °C. Characterization was conducted to determine particle size, elemental composition, functional groups, and optical properties. In addition, the PAI system was developed and evaluated by investigating the microphone frequency response under different arbitary function generator (AFG) frequencies, laser power stability, and the effects of duty cycle and modulation frequency. The system was first tested using agar-based phantoms and subsequently applied ex-vivo to BALB/c mouse salivary glands with variations in CDs injection volume and post-injection time. The results showed that the synthesized CDs had average particle sizes (approximately 1.5–6 nm), with a carbon content exceeding 94%, and functional groups (–OH and C=O) functional groups that contributed to colloidal stability. Optical characterization indicated that the CDs exhibited absorption extending to wavelength of 808 nm, making them suitable for photoacoustic excitation. The developed PAI system demonstrated stable performance, as evidenced by a linear microphone frequency response to variations in AFG frequency with a coefficient of determination close to unity (R² ? 1), laser power fluctuations less than 0.05 mW over 120-minute period, and a consistent response to duty cycle variations, reflected by corresponding changes in photoacoustic signal amplitude. The X–Y scanning system also exhibits good linearity. Phantom experiments identify an optimum modulation frequency in the range of 9–10 kHz with a duty cycle of 30%. The photoacoustic signal amplitude tended to increase with increasing CDs concentration, although the observed enhancement was relatively small (approximately 1 dBFS) and remained within the measurement deviation range. A homogeneous distribution of CDs was confirmed by histogram analysis and a coefficient of variation (CV) below 10%. Ex vivo experiments on BALB/c mouse salivary glands demonstrated a tendency toward increased photoacoustic signal amplitude following CDs injection compared to tissues without contrast agents. Variations in signal intensity were observed at different injection volumes and post-injection times, which may be associated with particle distribution within the tissue. In conclusion, the developed PAI system combined with soursop leaf-derived CDs demonstrates the potential of these nanomaterials as optical contrast agent for enhancing biological tissue imaging. However, further optimization is required to improve system sensitivity and overall performance of the imaging system.
