Sintesis dan Kompleksasi Senyawa Chemoprevention Curcumin Boron-4 (CCB-4) dengan Sorbitol, Fruktosa, dan N-metilglukamin, serta Uji Aktivitasnya Pada Sel Kanker Payudara Tipe Luminal

Kanker payudara subtipe luminal (ER+ dan/atau PR+) merupakan kasus yang dominan dan sering kali mengalami kendala resistensi terhadap terapi hormonal maupun agen kemoterapi standar. Senyawa turunan diboronokurkumin yaitu Chemoprevention Curcumin Boron 4 (CCB-4) menunjukkan potensi sitotoksik yang menjanjikan, namun masih memiliki keterbatasan yaitu kelarutannya dalam air yang rendah. Penelitian ini bertujuan untuk mengembangkan senyawa CCB-4 melalui kompleksasi dengan gula yaitu sorbitol (So), fruktosa (Fr), dan N-metilglukamin (MG) untuk meningkatkan kelarutan dan efektivitas sitotoksiknya serta mengevaluasi fenomena selulernya pada kanker payudara luminal T47D dan MCF-7.

Penelitian ini mencakup tahapan sintesis CCB-4 melalui kondensasi Claisen-Schmidt antara asam (4-formilfenil)boronat dan 1-metilpiperidin-4-on menggunakan katalis basa (NaOH), dilanjutkan dengan preparasi kompleks CCB-4-gula menggunakan metode pencampuran fisik. Karakterisasi kompleks dilakukan dengan menguji ukuran partikel, kelarutan senyawa, analisis gugus fungsional serta serapan maksimum UV-Vis. Evaluasi biologis CCB-4 dan kompleksnya dilakukan secara in vitro menggunakan uji MTT untuk menentukan indeks sitotoksisitas dan selektivitasnya. Evaluasi fenomena seluler pada T47D dan MCF-7 juga dilakukan mencakup penghambatan proliferasi sel, modulasi siklus sel, induksi mitotic catastrophe dan apoptosis. Kemampuan penghambatan proliferasi sel dievaluasi melalui uji persisten menggunakan metode MTT assay dan clonogenic assay dengan pewarnaan gentian violet 0,1%. Mekanisme penghambatan pertumbuhan sel dikaji lebih mendalam melalui analisis profil siklus sel. Pengamatan fenomena mitotic catastrophe juga dievaluasi melalui pewarnaan May-Grunwald Giemsa (MGG), sedangkan uji induksi apoptosis dilakukan menggunakan flow cytometry.

Hasil penelitian menunjukkan bahwa senyawa CCB-4 berhasil disintesis menggunakan katalis NaOH dengan rendemen sebesar 70,37 ?n kemurnian 97,91 %. Kompleksasi dengan sorbitol, fruktosa, dan N-metilglukamin berhasil meningkatkan kelarutan CCB-4 berturut-turut sebesar 13,25; 17,17; dan 25,14 kali lipat dibandingkan senyawa bebasnya. Kompleks CCB-4 dengan gula dapat meningkatkan aktivitas sitotoksik dan selektivitas dibandingkan CCB-4 bebas. Nilai IC50 berturut-turut 8, 6, 5, dan 5 ?M untuk CCB-4, CCB-4-So, CCB-4-Fr, dan CCB-4-MG pada sel T47D. Pada sel MCF-7, didapatkan IC50 yang lebih besar akibat pemberian senyawa CCB-4, CCB-4-So, CCB-4-Fr, dan CCB-4-MG yaitu 15, 8, 7, dan 9 ?M. Senyawa CCB-4 dan kompleksnya mampu menghambat proliferasi sel, menginduksi penghentian siklus sel pada fase G2/M, memicu mitotic catastrophe dan kematian sel melalui jalur apoptosis. Dari hasil penelitian ini dapat disimpulkan bahwa kompleksasi CCB-4 dengan gula efektif meningkatkan kelarutan dan potensi sitotoksiknya. Temuan ini menjadi dasar bagi pengembangan lebih lanjut kompleks CCB-4-gula sebagai kandidat agen antikanker payudara.

Luminal subtype breast cancer (ER+ and/or PR+) is the predominant form of the disease but often faces challenges regarding resistance to hormonal therapies and standard chemotherapeutic agents. A diboronocurcumin derivative, Chemoprevention Curcumin Boron 4 (CCB-4), demonstrates promising cytotoxic potential but is limited by low water solubility. This study aimed to modify CCB-4 through complexation with sugars specifically sorbitol (So), fructose (Fr), and N-methylglucamine (MG) to enhance its solubility and cytotoxic efficacy, and to evaluate its cellular mechanisms in T47D and MCF-7 luminal breast cancer cell lines.

The research involved synthesizing CCB-4 via Claisen-Schmidt condensation of 4-formylphenylboronic acid with 1-methylpiperidin-4-one using a basic catalyst (NaOH), followed by preparing CCB-4-sugar complexes by physical mixing. Complex characterization included assessments of particle size and morphology, as well as improvements in compound solubility. Biological evaluation of CCB-4 and its complexes was conducted in vitro using the MTT assay to determine cytotoxicity and selectivity indices. Cellular effects in T47D and MCF-7 cells were also evaluated, including inhibition of cell proliferation, cell cycle modulation, and induction of mitotic catastrophe and apoptosis. Cell proliferation inhibition was assessed using MTT and clonogenic assays (with 0.1% gentian violet staining). Mechanisms of growth inhibition were further investigated through cell cycle profile analysis. Mitotic catastrophe was observed by May-Grünwald-Giemsa (MGG) staining, and apoptosis induction was evaluated by flow cytometry. 

The research results demonstrate that the compound CCB-4 was successfully synthesized using NaOH as a catalyst, yielding 70.37% and achieving a purity of 97.91%. Complexation with sorbitol, fructose, and N-methylglucamine successfully increased the solubility of CCB-4 by 13.25, 17.17, and 25.14 fold, respectively, compared to the free compound. CCB-4-sugar complexes exhibited enhanced cytotoxic activity and selectivity compared to free CCB-4. The IC50 values ??were 8, 6, 5, and 5 ?M for CCB-4, CCB-4-So, CCB-4-Fr, and CCB-4-MG, respectively, in T47D cells. In MCF-7 cells, higher IC50 values ??were observed upon treatment with CCB-4, CCB-4-So, CCB-4-Fr, and CCB-4-MG: 15, 8, 7, and 9 ?M, respectively. CCB-4 and its complexes were capable of inhibiting cell proliferation, inducing cell-cycle arrest at the G2/M phase, and triggering mitotic catastrophe and cell death via the apoptotic pathway. In conclusion, complexation of CCB-4 with sugars effectively enhances its solubility and cytotoxic potential. These findings serve as the basis for further development of the CCB-4-sugar complex as a candidate anti-breast-cancer agent.
