Ekspresi protein bcl-2 pada karsinoma nasofaring

2005 | Tesis | PPDS I Patologi AnatomiLatar belakang : Protein bcl-2 salah satu famili protein yang menghambat apoptosis melalui jalur mitokondria, dan keseimbangan antara proliferasi sel dan kematian sell pada populasi tumor sebanding dengan besar dan stadium tumor. Tujuan : Mengetahui hubungan ekpresi protein bcl-2 dengan besar dan stadium pada penderita karsinoma nasofaring (KNF) sebelum dilakukan terapi. Cara penelitian : Pengecatan imunohistokimia dengan menggunakan antibodi monoklonal terhadap protein bcl-2 pada 47 sampel blok parafin penderita primer KNF. Metode yang digunakan standard streptavidin-biotin complex. Hasil : Ekpresi protein bcl-2 32 dari 47 kasus KNF (68,08%) ; 8 dari 10 kasus karsinoma tanpa keratinisasi dan 24 dari 37 kasus karsinoma tidak terdiferensiasi. Tidak ada hubungan bermakda antara kedua bentuk histologi. Ekpresi protein bcl-2 lebih tinggi pada penderita KNF stadium lanjut dibandingkan pada stadium dini. Kesimpulan : Hubungan bermakna ekpresi protein bcl-2 dengan stadium lebih lanjut penderita KNF.
Background : Bcl-2 protein is one of bcl-2 family protein which inhibits apoptosis through the mitochondria-dependent pathway, and the balance between cell proliferation, differentiation and cell death determines the cell numbers in a population, as well as the size and stage of a tumor. Aims : To investigate the association of expression bcl-2 protein with the size and stage of nasopharyngeal carcinoma (NPC) patients before therapeutic approach. Methods : Immunohistochemical staining using monoclonal antibodies using monoclonal antibody against bcl-2 protein was performed in 47 primary NPC with standard streptavidin-biotin complex technique. Results : The expressions of bcl-2 was detected in 32 of 47 cases of NPC (68,08%), consisted of 8 of 10 non keratizing carcinoma and 24 of 37 cases of undifferentiated carcinoma. There is no significant difference between two histological types. The expressions of bcl-2 protein in NPC patiens with more advance T and N staging were significantly higher than NPC patiens with more early T and N staging. Conclusion : This study indicate that the expression of bcl-2 protein is related to the late stage of NPC.
