This study developed a silver nanoparticle (AgNPs)-based colorimetric sensor system coated with ethylenediaminetetraacetic acid (EDTA) for the rapid, simple, and cost-effective detection of histamine in food products. AgNPs-EDTA were synthesized using ascorbic acid as a reducing agent and characterized using UV-Vis, FTIR, TEM, PSA-DLS, zeta potential, and stability tests. The histamine detection process was carried out by varying the pH, AgNPs-EDTA concentration, and interaction time to determine the optimal conditions. Additionally, to support method validation, tests for linearity, LOD and LOQ, selectivity, precision, and accuracy were conducted. These tests were performed using RGB digital image analysis and UV-Vis spectrophotometry.

EDTA-coated silver nanoparticles were successfully synthesized using ascorbic acid as a reducing agent. Characterization results using UV-Vis, FTIR, TEM, PSA-DLS, and zeta potential confirmed the formation of AgNPs stabilized by EDTA on the nanoparticle surface. The optimal conditions for histamine detection using AgNPs-EDTA were obtained at pH 7, an AgNPs-EDTA concentration of 400 µM, and an interaction time of 1 minute. The interaction between AgNPs-EDTA and histamine triggers nanoparticle aggregation, characterized by a shift in wavelength from 398 nm to 600 nm, resulting in a color change in the solution that forms the basis for colorimetric detection. The sensor exhibits a linear range of 250–1.000 µM; a limit of detection (LOD) of 3,59 µM and a limit of quantification (LOQ) of 11,96 µM in the UV-Vis method; an LOD of 3,65 µM and an LOQ of 12,15 µM in the RGB digital image analysis method; it is selective for histamine; the %RSD values for repeatability and reproducibility in the UV-Vis and RGB methods range from 1,08–1,93%, and the %recovery results range from 93,77%–102,81% in the UV-Vis method and 94,78–102,31% in the RGB digital image analysis method.
