Beef cattle are one of the most important livestock commodities contributing to the supply of animal protein for the community. Gunungkidul Regency, Special Region of Yogyakarta, is one of the regions with high population of a beef cattle and therefore has the potential to serve as a source of zoonotic transmission. One of the enteric bacteria that can be found in beef cattle is Salmonella sp., a pathogenic bacterium capable of causing salmonellosis in both animals and humans. Information regarding the occurrence of Salmonella sp. in beef cattle and its antibiotic resistance profile is essential for zoonotic disease control and the prudent use of antibiotics. This study aimed to determine the occurrence of Salmonella sp. and its antibiotic resistance profile in beef cattle in Gunungkidul Regency, Special Region of Yogyakarta. This study employed a cross-sectional design using 176 samples consisting of rectoanal mucosal swabs and fecal samples collected from 88 beef cattle. Isolation and identification of Salmonella sp. were performed using Buffered Peptone Water (BPW), Rappaport Vassiliadis Soya (RVS), and Xylose Lysine Deoxycholate Agar (XLD), followed by Gram staining, the Triple Sugar Iron Agar (TSIA) test, and the IMViC biochemical tests (Indole, Methyl Red, Voges–Proskauer, and Citrate). Antibiotic susceptibility testing was carried out using the Kirby–Bauer disk diffusion method against ciprofloxacin, penicillin, doxycycline, ampicillin, trimethoprim sulfamethoxazole, and streptomycin. Data were analyzed descriptively. The results showed that 4 of the 176 samples (2.27%) were identified as positive for Salmonella sp. based on isolation and identification procedures. Antibiotic susceptibility testing of the Salmonella sp. isolates revealed that 50% of the isolates were susceptible and 50% were intermediate to the antibiotics tested, while no resistant isolates were detected. These findings indicate that the occurrence of Salmonella sp. in beef cattle in Gunungkidul Regency was relatively low, and the isolates obtained remained highly susceptible to the antibiotics tested.
