Hard ticks of the genus Amblyomma are ectoparasites with the potential to act as vectors of various pathogens, making accurate species identification essential. This study aimed to identify a single Amblyomma hard tick collected from Kebumen, Central Java, based on morphological and molecular characteristics, and to analyze its phylogenetic relationship with reference isolates available in GenBank. Morphological identification was performed by examining the scutum, capitulum, eyes, festoons, coxae, and ornamentation pattern. Molecular identification involved DNA extraction, amplification of the mitochondrial cytochrome c oxidase subunit I (COI) gene using the universal primers LCO1490 and HCO2198, producing an amplicon of approximately 700 bp, followed by agarose gel electrophoresis, PCR product purification, DNA sequencing, and phylogenetic analysis using Molecular Evolutionary Genetics Analysis (MEGA) version 12 with the Kimura 2-Parameter (K2P) model and the Neighbor-Joining method. Morphological examination indicated that the tick possessed characteristics consistent with Amblyomma testudinarium. Genetic distance analysis showed that the Kebumen isolate had the closest genetic distance to a reference Amblyomma testudinarium isolate from China available in GenBank, with a genetic distance of 0.057 (5.7%). Phylogenetic analysis further demonstrated that the Kebumen isolate clustered within the same clade as the reference A. testudinarium isolate, with a bootstrap value of 100. Based on both morphological and molecular analyses, the hard tick isolate from Kebumen was identified as Amblyomma testudinarium.
