Indigofera tinctoria is a leguminous shrub commonly utilized for improving soil fertility, while its leaves are used as natural dyes and animal feed. The lignocellulosic biomass of I. tinctoria has potential as a second-generation bioethanol feedstock due to its structural carbohydrate fractions that can be converted into fermentable sugars. However, information regarding the chemical properties of I. tinctoria and its potential utilization as a bioethanol feedstock based on plant section and diameter classes remains limited. Therefore, this study aimed to determine the effects of diameter class and plant section on the chemical properties and reducing sugar yield as indicators of the potential utilization of I. tinctoria as a bioethanol feedstock.

Samples of 20-month-old I. tinctoria were collected from plot 17 of KHDTK Wanagama, Gunungkidul and classified into three diameter classes (small, medium, and large) and three plant section (stem, branch, and twig). The samples were ground to 40–60 mesh and analyzed for their chemical properties, including hot-water-soluble extractives, holocellulose, alpha-cellulose, and hemicellulose contents. The samples were then subjected to enzymatic hydrolysis to determine the hydrolysis rate and reducing sugar content using the DNS method.

The results showed that the interaction between diameter class and plant section had no significant effect on hot-water-soluble extractives, holocellulose, alpha-cellulose, hemicellulose, and reducing sugar content. However, the interaction significantly affected the hydrolysis rate, with the highest value of 13.20% obtained from the medium-diameter branch samples. The reducing sugar content was relatively uniform across all treatments, with an average value of 2.36 g/L. The results indicate that I. tinctoria biomass has potential as a bioethanol feedstock without requiring strict feedstock segregation based on plant section or diameter class.
