ABSTRACT
Soybean (Glycine max L.) is one of the strategic food crops and commodities with high economic value. One of the problems causing yield losses is leaf spot disease. The causal agents of leaf spot disease have been reported to be highly varied, particularly from the genera Cercospora, Colletotrichum, Fusarium, and Corynespora. The symptoms produced by these pathogens are similar to each other. This research aims to identify the fungal species that cause leaf spot disease in soybean plants based on morphological and molecular identification. From field sampling, 28 isolates were obtained from symptomatic leaf samples collected from Dlingo, Pleret, and Playen. The macroscopic and microscopic characteristics of the isolates showed diversity. Eight representative isolates were selected based on analysis using the UPGMA-Gower method, considering macroscopic and microscopic morphological characters as well as colony diameter on the 7th day, for Koch's postulate testing and molecular identification. Koch's postulates tests demonstrated that all eight representative isolates are pathogenic and induce leaf spot symptoms in soybean plant leaves. Molecular identification was performed using the Internal Transcribed Spacer (ITS) gene marker on the eight fungal isolates. Based on molecular identification, the eight isolates were identified as different fungal species Fusarium sp., Colletotrichum sp., Nigrospora sp., Diaporthe sp., Pestalotioid, and Macrophomina phaseolina
