Newcastle Disease (ND) remains a major threat to the poultry industry due to its high transmission rate and substantial economic impact. This study aimed to analyze the effectiveness of Promix®, which contains a combination of herbs (Curcuma domestica, Curcuma xanthorrhiza) and probiotics (Bifidobacterium, Lactobacillus, Saccharomyces) to enhance the humoral immune response of laying hens against ND. Promix® was used because the combination of herbs and probiotics within it works synergistically as an optimal immunomodulatory agent to stimulate the body's immune system. A total of 40 ISA Brown strain laying hens at 13 weeks of age were used as research subjects, divided into a control group and a Promix® treatment group. The ND booster vaccination was administered when the hens were 17 weeks old. The parameters measured were the ND antibody titer profiles using the Hemagglutination Inhibition (HI) test across three periods: pre-vaccination, 2 weeks post-vaccination, and 6 weeks post-vaccination. The results showed that in the pre-vaccination phase, the treatment group had a significantly higher mean titer and GMT (P<0>0,05). At six weeks post-vaccination, the mean titer of the treatment group actually decreased and was lower than that of the control group, although the difference was not significant (P>0,05). The administration of Promix® as a feed additive has a positive effect on increasing ND antibody titers through the HI test, wherein the immunity level is proven to remain above the minimum protective standard despite undergoing a natural decline at the sixth week.
