The root lession nematode Pratylenchus spp. is a plant parasitic nematode that can reduce the productivity of coffee plants (Coffea sp.). This study aims to determine the species of Pratylenchus spp. that attack coffee plants based on morphological, morphometric, and molecular identification. The study was conducted on coffee plantations in Sleman, Kulon Progo, Boyolali, and Magelang Regencies. Nematode extraction was carried out using the Whitehead tray method. Molecular identification was carried out using PCR techniques with primers D2A and D3B in the D2-D3 region of the 28S rDNA gene, followed by sequencing and phylogenetic analysis. The results showed that coffee plants attacked by Pratylenchus spp. experienced symptoms such as yellowing leaves, stunted growth, and root necrosis and decay. The highest population density was found in Sleman Regency, which was 446.67 individuals per 10 grams of roots and 413.33 individuals per 100 mL of soil. The results of morphological, morphometric, and molecular identification indicate that the nematodes found are identified as Pratylenchus coffeae and Pratylenchus speijeri.
