The little spiderhunter (Arachnothera longirostra) is a small bird belonging to the Nectariniidae family and is commonly found in Sumatra, Java, and Bali. Arachnothera longirostra is a bird that feeds on nectar from banana flowers and ginger plants, while most species in this genus feed on insects. This study aims to examine the morphology of the dorsal surface of the tongue using a Scanning Electron Microscope (SEM) and its histological structure via Hematoxylin and Eosin (HE) staining.

A total of four little spiderhunter were collected from the Special Region of Yogyakarta, regardless of sex, and used in this study. Species identification was conducted at the Animal Systematics Laboratory, Faculty of Biology, Gadjah Mada University. The use of animals and research procedures were approved by the Ethics Committee of the Faculty of Veterinary Medicine, Gadjah Mada University, under number 178/EC-FKH/int./2025. Sample preparation was performed under anesthesia, the birds were then decapitated, and tongue tissue samples were collected. The four samples were processed for analysis using SEM and Hematoxylin-Eosin staining. The stained sections were observed under a light microscope and photographed using an OptiLab Viewer.

Observations using SEM revealed characteristic features such as an apex divided into a central piece and two lateral pieces then forming a tubular structure, a corpus with a row of caudal lingual papillae, and a radix that appeared folded like an accordion in the resting state. HE staining results showed that the entire region was covered by a keratinized layer, with the apex region consisting solely of a tubular keratinized epithelial layer, while the corpus and radix regions had a more complex structure comprising the mucosal epithelial lamina, the mucosal lamina propria, and the muscularis lamina.
