Dendrobium is one of the largest genera in the family Orchidaceae, comprising approximately 1,600 species. This genus exhibits an epiphytic habit and is distributed throughout tropical and subtropical regions of Asia. Studies on the anatomical characteristics of plants, particularly Dendrobium spp., remains limited in Indonesia. Meanwhile, anatomical information is essential for accurate taxonomic identification. This study aimed to obtain anatomical and histochemical data on the roots and stems of Dendrobium anosmum Lindl. and Dendrobium spathilingue J.J.Sm. Hand-sectioning was used for anatomical and histochemical analyses of the stems, while the paraffin embedding method was employed for root anatomical and histochemical analyses. Histochemical analyses included the detection of starch, lipids, lignin, proteins, and alkaloids using Lugol’s iodine (IKI), Sudan Black B, phloroglucinol-HCl, Coomassie Brilliant Blue, and Dragendorff’s reagent, respectively. The results revealed that the root anatomy consisted of velamen, exodermis, cortex, endodermis, and stele. Differences were observed in the velamen structure, which appeared circular in D. anosmum and irregular in D. spathilingue. In addition, the exodermal and endodermal cell walls of D. spathilingue exhibited thicker wall thickenings. Anatomically, the stem was composed of a cuticle, epidermis, hypodermis, ground tissue, and vascular bundles. Calcium oxalate crystals and folded water-storage cells were found in the ground tissue of D. anosmum. Histochemical tests showed positive reactions for proteins, lipids, lignin, and starch in the roots and stems, as well as alkaloids in the stems of both Dendrobium species.
