sebesar 6,38 a.u./Ų.
A computational chemistry study was conducted on the structural modeling of chitosan-based Molecularly Imprinted Polymer (MIP) using paclobutrazol
(PBZ) and gibberellic acid (GA3) as template molecules using the Density Functional Theory (DFT) approach. This study aimed to obtain the most stable MIP cavity model, determine the types and characteristics of non-covalent interactions, and analyze the interaction energy of chitosan-based MIP-template complexes to evaluate the stability of the controlled-release system for plant growth regulators.
The study began with computational method validation on PBZ, GA3, and chitosan dimer structures using DFT/B3LYP-D3BJ with def2-SVP, def2-TZVP, and def2-TZVP(-f) basis set variations. The validation results showed that def2-TZVP(-f) could be used as a representative method for MIP modeling. The MIP models were constructed from two chains of chitosan pentamer and hexamer connected by glutaraldehyde as a crosslinking agent in parallel and antiparallel orientations. Each MIP-template complex was optimized using ORCA 6.1.1, and its cavity volume was analyzed using Molovol. The interaction energy was calculated through single point energy calculations with the CPCM water solvent model. Non-covalent interactions were analyzed using the IGMH method through Multiwfn and visualized using VMD. The results showed that the hexamer_A model was the most stable chitosan- based MIP model for PBZ, with an interaction energy of -130.30 kJ mol?¹ and cavity volume of 288.33 ų, supported by the formation of a strong H117…N300 hydrogen bond with an IBSIW value of 3.30 a.u./Ų. For the GA3 template, the hexamer_B model showed the highest stability, with an interaction energy of -192.24 kJ mol?¹ and a cavity volume 456.80 ų, indicated by the H343…N173 hydrogen bond with an IBSIW value of 6.38 a.u./Ų. Based on these results, MIP stability was influenced by cavity volume compatibility, template orientation, active group distribution, and the strength of the non-covalent interactions formed. Therefore, hexamer_A and hexamer_B are the most promising chitosan-based MIP models for PBZ and GA3 binding, respectively.
