Oil palm (Elaeis guineensis Jacq.) is one of Indonesia’s most important plantation crops and plays a crucial role in meeting global vegetable oil demand. The increasing demand for palm oil has highlighted the need to develop superior oil palm planting materials capable of enhancing productivity without expanding cultivated land areas. One approach is genetic improvement through induced polyploidization as a plant breeding strategy to generate superior genotypes with greater productivity potential. In this study, in vitro polyploidization was induced using Bio-Catharanthine, a natural antimitotic agent derived from Catharanthus roseus. The treatments consisted of a control (1C) and nine treatment combinations comprising three Bio-Catharanthine concentrations, namely 1.5% (8), 3% (9), and 4.5% (10), combined with three induction durations of 24 hours (A), 48 hours (B), and 72 hours (C). This study aimed to analyze the effect of polyploidization induction on phenotypic characteristics and ploidy level in oil palm callus. Phenotypic analysis included morphological characteristics such as color and texture of the callus and anatomical characteristics such as the cell number and cell area, while genotypic analysis is performed by measuring ploidy level using the flow cytometry method. Anatomical data on cell number and area were analyzed using the Kruskal–Wallis test, Two Way ANOVA, and Scheirer–Ray–Hare (SRH) test using RStudio software. The results showed that Bio-Catharanthine treatment induced browning in callus tissues under specific treatment conditions. In addition, increasing the concentration and induction time decreased the number of cells and increased the area of oil palm callus cells. Treatments 10C (4.5%, 72 hours), 10B (4.5%, 48 hours), and 9C (3%, 72 hours) were the most effective in increasing the size of oil palm callus cells compared to other treatments. However, Bio-Catharanthine treatment did not induce changes in ploidy levels in oil palm callus. This was demonstrated by the main peak of the flow cytometry histogram for all treatments being similar to the control, with the major peak remaining within the fluorescence intensity range of 4 × 10³ to 5 × 10³. Thus, Bio-Catharanthine induction affects phenotypic characteristics, but has not been effective in inducing changes in ploidy levels in oil palm callus.
