Escherichia coli (E. coli) are bacteria commonly associated with zoonotic diseases and antibiotic resistance. E. coli spreads easily in the environment, including through contaminated water. Wastewater generated by slaughterhouses is a potential source of E. coli contamination in the environment if not properly treated. This study aims to evaluate the potential for E.  coli contamination of the environment at three ruminant slaughterhouses in the Special Region of Yogyakarta, which are the Giwangan Slaughterhouse, the Mancasan Slaughterhouse, and the Kentungan Slaughterhouse. Samples were collected from the influent channel (before treatment), the treatment pond, and the effluent channel (after treatment). Bacterial enumeration was performed using the pour-plate method on TBX medium, and counts were calculated using the formula based on ISO 7128:2007. The results showed that the number of E. coli in wastewater in the influent and effluent channels at the Giwangan and Kentungan slaughterhouses decreased, from an average of bacteria count from 8.7×10? CFU/ml to 3.3×10² CFU/mL of E. coli in the effluent at the Giwangan Slaughterhouse and from an average of 7.2×10? CFU/ml to 6.9×10? CFU/ml E. coli in the effluent at the Kentungan Slaughterhouse. Meanwhile, at the Mancasan Slaughterhouse, there has been an increase in the number of E. coli in the discharged wastewater, specifically from 9.4×10? CFU/ml to 2.6×10? CFU/ml in the infiltration pond, which serves as the discharge point. The results of this study highlight the importance of slaughterhouse wastewater treatment in reducing E. coli in wastewater.
