Dendrobium is the second-largest orchid genus after Bulbophyllum Thouars within the family Orchidaceae. Dendrobium spathilingue belongs to section Calcarifera, whereas Dendrobium anosmum is classified under section Dendrobium. Species within the genus Dendrobium exhibit diverse pollination strategies, in which the floral sepals and petals, together with characteristics such as color, texture, volatile compounds, and epidermal derivatives including papillae, function as visual and chemical attractants for pollinators. However, information regarding the anatomical and histochemical structures of the sepals and petals, particularly in D. spathilingue and D. anosmum remains limited. Therefore, this study was conducted to analyze the anatomical and histochemical characteristics of these two species and to examine their potential relationship with pollination functions. Anatomical observations were performed using hand-sectioning techniques to investigate paradermal structures, while paraffin-embedded transverse sections of sepals and petals were prepared for anatomical and histochemical analyses. Histochemical tests were conducted to detect lipids, proteins, starch, lignin, and alkaloids using Sudan Black B, Coomassie Brilliant Blue, IKI, phloroglucinol-HCl, and Dragendorff’s reagent, respectively. The results revealed distinct morphological differences between the sepals and petals of D. spathilingue and D. anosmum, particularly in organ shape, apex morphology, and coloration. Anatomically, D. spathilingue was characterized by the presence of hypodermis and osmophores in the dorsal sepals, which were absent in D. anosmum, whereas D. anosmum was characterized by conical papillae on the lateral sepals and lower epidermal cells with elongated and conical shapes on the petals, which were not observed in D. spathilingue. Histochemically, both species showed relatively similar patterns, including the presence and distribution of lipids, proteins, and alkaloids, while starch and lignin were absent. These similarities suggest that the sepals and petals may function in metabolite storage and contribute to pollination rewards involved in attracting pollinators.
