The orange-spotted grouper (Epinephelus coioides) is a predatory fish characterized by a yellowish-brown body coloration with orange spots distributed throughout its body. This species inhabits the Indo-Pacific waters and is subject to exceptionally high market demand, resulting in increasingly intensive harvesting, including through destructive methods such as blast fishing and the use of active chemical substances such as methomyl. The gills represent the primary organ of the respiratory system in fish and serve as the first anatomical structure exposed to waterborne pollutants; however, the histological architecture of the respiratory system of the orange-spotted grouper under normal conditions and following methomyl exposure has not yet been reported. This study aimed to examine the histological and mucin profiles as glycoproteins of the respiratory organ of the orange-spotted grouper under normal conditions and methomyl-induced toxicity. Six adult specimens were divided into a control group of three individuals and a treatment group of three individuals exposed to 25% methomyl toxin. The treatment group was exposed to 1 mg of 25% methomyl in seawater for 30 minutes, whereas the control group underwent anesthesia by cold temperature immersion. Samples were subsequently fixed and processed into histological preparations stained using Hematoxylin-Eosin (HE) to observe tissue architecture and Alcian Blue-Periodic Acid Schiff (AB-PAS) to examine the mucin profile in both groups. Observations were conducted using light microscopy and documented with Optilab Viewer. Data analysis was performed descriptively to characterize histopathological changes (HE) and semi-quantitatively (AB-PAS) using the systematic random sampling method to describe the mucin profile at each zone in both groups. The results demonstrated histopathological alterations including epithelial lifting, edema, absence of secondary lamellae, telangiectasia, hyperplasia, and necrosis in the treatment group. Furthermore, a significant difference in mucin distribution was observed between groups, concentrated in the apical zone, with a shift in dominant mucin type from mixed mucins in the control group to acidic mucins in the treatment group.
