asam amino gen ERECTA.
Douglas berukuran mini.
Orchids are one of the
largest flowering plant family in Indonesia and have high economic value as
ornamental plants. One of the most widely cultivated species the orchid Vanda
Douglas (Papilionanthe Miss Joaquim) is known for its large flowers
and high commercial value. However, more compact or miniature plants are
increasingly in demand because they are more suitable for decoration and indoor
cultivation with limited space. One approach that has the potential to support
the development of miniature orchids is genome editing using CRISPR/Cas9
technology. As a basis for applying this technology, homology analysis of the
target gene is required to identify the level of sequence similarity, protein
characteristics, and candidate target regions or protospacer sequences in the ERECTA
gene which have the potential to be used in genome editing. Gene ERECTA known
to play a role in regulating the growth and development of plant organs, while
mutations in homologous genes in Arabidopsis thaliana was found to cause
shortening of flower stems and changes in plant morphology. Therefore, this
study aims to analyze the homology of gene sequences ERECTA gene homologous
to orchid Vanda Douglas work focused on nucleotide and amino acid
sequences, analysis of protein motif characteristics, and analysis of potential
target sites for CRISPR/Cas9 based genome editing. The research was conducted
through observation of plant morphological and anatomical characteristics,
followed by analysis.in silico to the nucleotide and amino acid
sequences of gene ERECTA. The results of the study showed that the gene ERECTA
homologues in Vanda Douglas have domains Leucine-Rich Repeat (LRR)
with conservative amino acid sequences DARGNKLNGPIPRSFKNL then GNKL protein motif and GTIP which is
thought to play a role in the function of the ERECTA protein. In addition, this
study successfully analyzed candidate target sites for CRISPR/Cas9-based genome
editing with sequences GCACGAGTAACAAGTTAAATGG. The results of this study are
expected to form the basis for further research on the application of genome
editing to support the development of orchid plants Vanda Douglas to
creation mini-orchid.
