Hard ticks (Ixodidae) are ectoparasites that commonly infest dogs and act as vectors for various pathogens affecting both animal and human health. One of the most commonly found tick species is Rhipicephalus sanguineus, which has a wide distribution in tropical and subtropical regions. Conventional tick identification based on morphological characteristics has limitations due to similarities among species; therefore, a more accurate molecular approach is required. One molecular method that can be used is identification based on the mitochondrial DNA marker of the 16S rRNA gene. This study aimed to identify dog ticks collected from Sleman, Special Region of Yogyakarta, based on morphological and molecular characteristics using the 16S rRNA gene and to analyze their phylogenetic relationship with isolates from various countries. Tick samples were collected from Sleman, Special Region of Yogyakarta. DNA isolation was performed using the Thermo Scientific GeneJET Genomic DNA Purification Kit. The isolated DNA was amplified using the Polymerase Chain Reaction (PCR) technique with 16S+1 and 16S-1 primers, resulting in an amplicon size of 455 bp. The samples were subsequently sequenced at the Integrated Research and Testing Laboratory (LPPT), Universitas Gadjah Mada. Sequence analysis was conducted using MEGA version 12. The analyses included nucleotide differences, nucleotide composition, transition/transversion ratio, genetic distance estimation, and phylogenetic tree reconstruction. Genetic distance analysis using the Kimura 2-parameter method showed a value of 0.01 compared to the Taiwan isolate of Rhipicephalus sanguineus. Phylogenetic tree reconstruction based on the 16S rRNA gene using the Neighbour-Joining method demonstrated a close genetic relationship with the Taiwan isolate, supported by a bootstrap value of 100, indicating a high level of genetic similarity. It can be concluded that the tick isolate from Sleman was identified as Rhipicephalus sanguineus and showed close genetic relatedness to the Taiwan isolate
