Indonesia provides various regions suitable as habitats for wild silkworms of the genus Antheraea (Hübner, 1819), with a tropical climate and supported by the availability of humid heterogeneous forests, such as in the Special Region of Yogyakarta (DIY). Studies regarding the genetic diversity of wild silkworms in DIY have never been conducted. Research related to the genetic diversity of wild silkworms using a more comprehensive COI gene approach is expected to serve as a basis for conservation efforts of Antheraea. Therefore, this study aimed to determine the genetic diversity of wild silkworms in DIY using the COI gene. This research employed the PCR method with universal COI gene primers through several stages, including sampling, DNA extraction, DNA quality and quantity testing, DNA amplification, electrophoresis, gel documentation, and sequencing. The obtained data were then analyzed using the programs GeneStudio, BLAST, MEGA, and DnaSP. The results showed that the sampled Antheraea can be found in Muntingia calabura tree around DIY between elevation range from 20 to 450 meters above sea level. Furthermore, the Antheraea species identified in DIY was Antheraea jana. However, no genetic diversity was found in the sampled population in DIY, indicated by the presence of only one haplotype along the 558 bp fragment with 0 variable sites and 0 parsimony-informative sites. Subsequently, the haplotype diversity and nucleotide diversity values were both 0.000. The genetic distance among samples was 0.00. All these results shows that the samples have similar genetic identity. Phylogenetic analysis indicated that Antheraea jana from DIY formed a clade with Antheraea jana from Sumatra Barat, Java Barat, and Java Timur within the same species branch with a bootstrap value of 100 and formed a different clade with the same species from NTB and Bali. This study is expected to contribute as foundational data for the development of a COI gene database related to wild silkworm conservation of Antheraea in Indonesia, particularly in the Special Region of Yogyakarta.
