Phytotherapy has emerged as a promising alternative to synthetic drugs by utilizing bioactive compounds from medicinal plants with fewer side effects. Among medicinal plants with pharmaceutical potential, okra (Abelmoschus esculentus) is rich in bioactive compounds and has been reported to exhibit various pharmacological activities, including anticancer, cardioprotective, etc. To support the sustainable production of these valuable metabolites, plant tissue culture provides a scalable and efficient in-vitro platform for pharmaceutical applications. This study aimed to establish and evaluate a sequential experimental pipeline for okra leaf callus culture, comprising of optimization of explant surface sterilization, assessment of callus induction through callogenesis stages and induction frequency, and metabolite profiling of culture medium exudates using LCHRMS. The methodology included ex-vitro cultivation of okra plants, leaf sterilization optimization, in-vitro callus culture, culture medium extraction, and LC-HRMS analysis. Results showed that the optimized sterilization protocol consisted of immersion in a 1.5 ppm bactericide-fungicide solution for 20 minutes, followed by treatment with 0.6% NaOCl-Tween 20 for 3 minutes, 70% alcohol for 30 seconds, and mechanical removal of leaf trichomes. Using this optimized protocol, three sequential stages of callogenesis were observed, namely wound response, cell division, and callus formation, with 68% of explants reaching the final pluripotency stage. Furthermore, LC-HRMS analysis identified 32 compounds belonging to 9 metabolite groups, predominantly primary metabolites such as amino acids and organic acids. Overall, this study successfully established an effective protocol for okra leaf callus culture through optimized surface sterilization and demonstrated a high callogenesis success rate. In addition, the metabolite profile of culture medium exudates indicates their potential as biomarkers for callus growth monitoring. Future studies should investigate elicitor treatment to enhance secondary metabolite production and a comparison between different plant growth regulators (PGRs) to improve callus organogenesis and metabolite accumulation
