Female bamboo (Bambusa balcooa) is a member of the Poaceae family that originates from India but is now widely distributed throughout Indonesia. This plant is known for various health benefits, including antidiabetic, antioxidant, anti-inflammatory, and gastroprotective properties. However, the utilization of B. balcooa in the health sector has mainly focused on its shoots. The leaves of B. balcooa are known to contain various phytochemicals, such as flavonoids, alkaloids, oxylipins, and phenolic compounds. These compounds have the potential to act as antioxidants that can reduce the levels of free radicals in the body. Excessive free radical accumulation may lead to various health problems, including inflammation. This study aimed to evaluate the antioxidant activity of B. balcooa leaf extract in vitro and to investigate its potential as an anti-inflammatory agent in silico. Antioxidant activity was assessed using the DPPH assay and the Total Antioxidant Capacity (TAC) assay based on the Ferric Reducing Antioxidant Power (FRAP) method. The anti-inflammatory potential was evaluated in silico through molecular docking using Cyclooxygenase-2 (COX-2; PDB ID: 5IKR) as the target protein. The results of the DPPH and TAC assays demonstrated that B. balcooa leaves possess good antioxidant activity. The ethyl acetate fraction exhibited the best antioxidant performance, with an IC50 value of 18,7 ppm (categorized as very strong) in the DPPH assay and a TAC value of 91,77 ?mol/mL at concentration of 5.000 ppm in the TAC assay. B. balcooa leaves also showed potential as an anti-inflammatory agent through the inhibition of COX-2, which may be attributed to the presence of oxylipin and phenolic compounds. Based on the molecular docking results, 12-oxo-phytodienoic acid (OPDA) demonstrated the strongest binding affinity toward COX-2, with a binding energy of -7,3 kcal/mol. Although this value was lower than that of the co-crystallized ligand (-8,0 kcal/mol), OPDA exhibited the highest binding affinity among all identified compounds, indicating its potential as a candidate COX-2 inhibitor.
